摘要
目的:观察不同剂量的蜂针对佐剂诱导的类风湿性关节炎大鼠成纤维滑膜细胞活力的影响,探讨蜂针治疗类风湿性关节炎的可能机制。方法:建立大鼠佐剂性类风湿性关节炎动物模型,将Wistar大鼠随机分为空白组、模型组和单蜂1 min组、双蜂1 min组、三蜂1 min组、单蜂30 s组,观察各组足底厚度以及HE染色的病理变化,同时关节滑膜取材,采用MTT法检测成纤维样滑膜细胞增殖情况。结果:足底厚度比较,1 min蜂针组较模型组以及其他蜂针剂量组别低(P<0.01);HE病理形态学评分,模型组病理形态学改变评分较各蜂针治疗组高(P<0.05);MTT检测成纤维滑膜细胞活力中,24 h时间点开始出现,1 min蜂针组抑制成纤维细胞增殖较模型组显著(P<0.05),48 h较24 h时间点各组差异更显著。结论:适当剂量的蜂针能减轻类风湿性关节炎大鼠足底厚度,降低病理形态学评分,其机制可能跟蜂针抑制成纤维样滑膜细胞增殖活力有关。
Objective:To observe the effect of different doses of bee venom therapy on fibroblast-like synoviocytes in rats with adjuvant arthritis and explore the possible mechanism of different doses of been venom therapy in the treatment of rheumatoid arthritis.Methods:The adjuvant arthritis rat model was established.Wistar rats were randomly divided into normal control group,model group,single bee 1 min group,double bees 1 min group,three bees 1 min group,single bee 30 s group.Foot thickness and pathological scores were observed and activity of FLS proliferation in RA rats was detected by MTT assay.Results:The foot thickness of single bee 1 min group of rats was significantly lower than that of the model group(P〈0.01),the pathological scores corresponds lower than that of the model group(P〈0.05),and the FLS proliferation in single been 1 min group was lower than that of the model group in 24 h(P〈0.05),and that at 48 h was obvious.The inhibition of FLS proliferation in single bee 1 min group was better than other treatment groups.Conclusion:Appropriate dose of venom therapy can relieve toe swelling of rheumatoid arthritis in rats and alleviate the pathological changes,which may relate to the mechanism of RA that inhibits activity of FLS proliferation.And the single bee 1 min may be the optimal therapeutic dose of rheumatoid arthritis.
