摘要
目的:观察一贯煎对肝癌表皮生长因子受体(epidermal growth factor receptor,EGFR)信号通路及其下游侵袭转移相关蛋白细胞外调节蛋白激酶(extracellular regulated protein kinases,ERK)、增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)、细胞间黏附分子-1(intercellular cell adhesion molecule-1,ICAM-1)表达的影响。方法:雄性KM小鼠50只,随机均分为模型组,环磷酰胺(cyclophosphamide,CTX)组,一贯煎高剂量组、一贯煎中剂量组、一贯煎低剂量组。腋下注射H22细胞建立小鼠荷瘤模型,一贯煎高剂量组、一贯煎中剂量组、一贯煎低剂量组造模前两周开始用药,剂量分别为46 g·(kg·d)^(-1)、23 g·(kg·d)^(-1)、11.5 g·(kg·d)^(-1),CTX组造模后开始给药,剂量为0.05 g·(kg·d)^(-1),模型组造模后灌胃等体积生理盐水,造模后继续给药2周。处死各组小鼠取瘤称质量,蛋白电泳法检测EGFR、p-ERK、PCNA及ICAM-1蛋白的表达。结果:与模型组比较,一贯煎各剂量组及CTX组均能显著降低瘤体质量,差异有统计学意义(P<0.01);一贯煎各剂量组与CTX组比较,瘤体质量均明显降低,差异有统计学意义(P<0.01)。与模型组比较,一贯煎高剂量组可显著降低EGFR及PCNA蛋白表达,一贯煎高剂量组及一贯煎中剂量组可显著降低p-ERK及ICAM-1蛋白表达,差异有统计学意义(P<0.05)。结论:一贯煎可能通过调控EGFR信号通路及其下游侵袭转移相关蛋白p-ERK、PCNA、ICAM-1蛋白表达,发挥抑制荷H22小鼠肝癌细胞侵袭转移的作用。
Objective:To observe the effect of Yiguan Jian on the growth of liver cancer and its effects on the EGFR signaling path way which regulated the expression of invasion and metastasis related proteins ERK,PCNA,ICAM-1.Methods:50 male KM mic were randomly divided into model group,chemotherapy group,and Yiguan Jian high,middle,low dose group.H22 tumor cells wa axillary injected to established bearing mice model,consistently intragastric administrated with Yiguan Jian high dose 46 g·(kg·d)-1,medium dose 23 g·(kg·d)-1and low dose 11.5 g·(kg·d)-1in traditional Chinese medicine treatment group tw weeks before modeling,CTX was administrated in chemotherapy group after modeling,0.05 g·(kg·d)-1,the mice in mode group was administrated with the same volume of normal saline,and continuous medication for 2 weeks after modeling.Killed th mice and weighed tumor tissue,detected the protein expression of EGFR,p-ERK,PCNA,ICAM-1 by westerning blot.Results:Th results showed that compared with the model group,there was a significant reduction the quality of tumors in the high,medium and low dose Yiguan Jian group and CTX group(P〈0.01),and there was significant difference between the each dosage Yiguan Jian group and CTX group(P〈0.01).Compared with the model group,high dose of Yiguan Jian group could significantly reduce EG-FR and PCNA protein expression,high dose and medium dose could significantly reduce protein expression of p-ERK and ICAM-1,there were significant differences between them(P〈0.05).Conclusion:It is possible to regulate the expression of PCNA,pERK,ICAM-1 protein by regulating the EGFR signaling pathway and its downstream invasion and metastasis associated proteins.The effect of inhibiting the invasion and metastasis of hepatocellular carcinoma cells in H22 tumor-bearing mice.
