建立多囊卵巢综合征胰岛素抵抗大鼠模型及其生殖内分泌改变的实验研究 被引量:6

摘要 目的建立大鼠多囊卵巢综合征(PCOS)胰岛素抵抗(IR)动物模型,探究其卵巢病理变化及生殖内分泌异常。方法将120g左右的雌性SD大鼠随机分为空白对照组,来曲唑组,来曲唑+高脂组,每组10只。来曲唑组每日常规饲料喂养并灌服来曲唑0.5%CMC-Na溶液1mg·kg-1,来曲唑+高脂组每日采用高脂饲料喂养并灌服来曲唑0.5%CMC-Na溶液1mg·kg-1,空白对照组每日自由进食饮水。观察动情周期、体重和卵巢组织学变化,测定糖耐量(OGTT)、空腹血糖(FPG)和空腹胰岛素(FINS)浓度,并计算胰岛素抵抗指数(HOMA-IR),检测血清性激素促黄体生成素(LH)、卵泡刺激素(FSH)、雌二醇(E2)、孕酮(P)、睾酮(T)。结果来曲唑组和来曲唑+高脂组大鼠均失去规律动情周期,且卵巢呈多囊样改变,卵巢闭锁卵泡增多,闭锁卵泡直径增大,颗粒细胞层数明显减少,黄体数量减少。与对照组比较,来曲唑+高脂组大鼠体重增长幅度增加(P<0.01),FINS升高(P<0.01),HOMA-IR升高(P<0.05)。血清性激素LH、T、LH/FSH升高,P、E2降低。结论来曲唑配合高脂饮食可成功诱导多囊卵巢综合征胰岛素抵抗大鼠模型。 OBJECTIVE To establish rat models for the study of polycystic ovary syndrome-insulin resistance( PCOS-IR),and evaluated ovarian pathological changes and reproductive endocrine abnormalities. METHODS120 g female SD rats were randomly divided into 3 groups: control group,letrozole group,letrozole + high-fat group. The rats in the letrozole group were fed with a normal diet and 1 mg·kg-1 of letrozole 0. 5% CMC-Na. The high-fat diet group was fed with high-fat diet and 1 mg·kg-1 of letrozole 0. 5% CMC-Na,the control group had free access to food and water. Estrous cycle,weight and ovarian morphology were observed,glucose tolerance test( OGTT),blood fasting plasma glucose( FPG),blood fasting insulin( FINS) and homeostasis model of assessment for insulin resistance index( HOMA-IR) were determined,and serum sex hormone luteinizing hormone( LH),follicle stimulating hormone( FSH),estradiol( E2),progesterone( P),testosterone( T) were detected in all the groups. RESULTS Compared with control group,the letrozole group and letrozole + high-fat group showed polycysic ovaries with irregular estrous cycle,increased number and diameter of atretic follicle,decreased layers of granulosa cells and few corpora lutea. The weight increased were singnificantly higher than the contorl( P 0. 01); above two groups also showed higher FINS concentration( P 0. 01) and increased HOMA-IR( P 0. 05); the serum hormone level: LH,T,LH/FSH in letrozole experimental group were significantly higher than the control,and E2 and P decreased significantly. CONCLUSION Letrozole with high-fat diet can successfully induce typical metabolic phenotypes of insulin resistance in polycystic ovary syndrome rats.
