摘要
从龙岩市某污水处理厂的活性污泥中富集分离纯化得到一株具有较高脱氮能力的异养硝化-好氧反硝化菌,命名为LT-11。通过菌落形态、生理生化和16S r DNA序列分析,LT-11初步鉴定为Pseudomonas putida。通过单因素实验考查碳源、pH、温度及溶解氧对该菌株的脱氮性能影响。结果表明:菌株LT-11最适碳源为琥珀酸钠,pH为7~7.5,温度为25~30℃,摇床转速180 r/min,该菌株氨氮去除率达90%以上。同时该菌株具有良好的好氧反硝化能力,24 h硝酸盐氮(初始浓度50mg·L-1)去除率接近80%,且没有亚硝酸盐氮积累。
A heterotrophic nitrification and aerobic denitrifying bacteria with high nitrogen removal capacity was enriched and purified from activated sludge of a se'wage treatment plant in Longyan City, which 'was named LT - 11. According to colony morphology,physiological biochemistry and 16S rDNA sequence analysis, the strain wTas identified as Pseudomonas putida. The effects of carbon source,pH,temperature and dissolved oxygen on the nitrogen removal of the strain wTere investigated by single factor experiments. The results showed that the removal rate of ammonia nitrogen of strain LT-11 reached above 90%,'when the optimum carbon source was sodium succinate,pH value 'was 7 ?7 .5 ,temperature 'was 25 ?30 尤 and shaking speed 'was 180 r/min. At the same time,the strain had good aerobic denitrification ability. The removal rate of nitrate nitrogen (initial concentration 50 mg? L 1 ) 'was close to 80% 'within 24 h,and no nitrite accumulation 'was observed.
