摘要
目的研究长链非编码RNA(long non-coding RNA,lncRNA)H19在食管鳞癌细胞中的作用与功能。方法运用实时荧光定量PCR(Real time quantitative polymerase chain reaction,qRT-PCR)检测5种食管鳞癌细胞系中lncRNA H19本底表达,用RNA干扰技术转染H19小干扰RNA(Small interfering RNA,siRNA)至食管鳞癌细胞KYSE30;实验分3组:空白对照组(空白载体组)、阴性对照组(随机序列组)和实验组(H19干扰组)。用噻唑蓝比色法(MTT)、流式细胞术(Flow Cytometry,FCM)、细胞划痕、Transwell侵袭实验检测lncRNA H19对食管鳞癌细胞增殖、凋亡、迁移及侵袭能力的影响。结果 5种食管鳞癌细胞系中,KYSE30细胞lncRNA H19表达量最高,其次分别是Eca109、KYSE150、EC9706和KYSE450细胞。实验组敲低KYSE30细胞中lncRNA H19的表达(P<0.05),细胞增殖受到抑制(P<0.05),细胞凋亡增加(P<0.05),细胞迁移距离明显减小(P<0.05),细胞侵袭能力明显减弱(P<0.05),与空白对照组和阴性对照组相比,差异有统计学意义。结论 lncRNA H19的表达能够促进食管鳞癌细胞的增殖、迁移和侵袭,并抑制细胞凋亡,食管癌中lncRNA H19发挥促进细胞恶性增殖及浸润、转移的癌基因作用,实验结果为今后可能用于临床诊断及治疗奠定了基础。
Objective To investigate the role of lncRNA H19 in esophageal squamous cell carcinoma(ESCC).Methods The expressions of lncRNA H19 in 5 ESCC cell lines were detected by qRT-PCR.H19 siRNA was transfected into KYSE30 cell to knockdown the expression of lncRNA H19,and the study was divided into the normal control,nagetive control and experimental groups.The effects of H19 on cell proliferation,apoptosis,migration and invasion were investigated by MTT,FCM,Wound-healing and Transwell invasive assay.Results The expression of lncRNA H19 was high in 5 ESCC cells.The knockdown expression of H19 in KYSE30 cellssuppressed cell proliferation(P <0.05),migration(P <0.05)and invasion(P <0.05),meanwhile accelerated cell apoptosis(P <0.05).Conclusion lncRNA H19 plays a role of oncogene in ESCC,thus probably promotes ESCC malignant proliferation and distant metastasis.
