摘要
目的针刺急性期脑出血模型大鼠百会透曲鬓穴,观察JNK通路被抑制后其下游通路蛋白pSTAT3的表达变化及头针对脑组织是否起到保护作用。方法选用雄性SD大鼠84只随机分为空白组与假手术组各6只,模型组、针刺组(模型+针刺组)与SP600125(模型+抑制剂组)各24只;造自体血脑出血模型,选择针刺百会穴透曲鬓穴的方法,在头部一针贯穿顶额颞三区。于造模成功后针刺,在6 h,1 d,3 d,7 d 4个时间点取材,采用免疫组化分析法(IHC)观察脑组织内p-STAT3的阳性细胞灰度值。结果针刺百会透曲鬓穴可以减轻模型动物神经缺损症状,提高评分。p-STAT3在大鼠脑出血后6 h表达开始增多,1 d表达相对最多,3 d达高峰后迅速减少,7 d仍有表达。针刺组各时间点阳性细胞灰度值均比模型组降低,具有显著差异。针刺组与SP600125组比较差异有统计学意义。结论针刺百会透曲鬓穴能够减轻大鼠神经功能缺损程度评分,并降低p-STAT3蛋白的表达,表明针刺在脑出血的治疗中起到了脑保护作用。
Objective: To observe the expression variation of JNK pathway's downstream of molecule-p-STAT3, after JNK pathway inhibition by acupuncture penetrating Baihui (GV20) and Qubin (GB7) on acute cerebral hem- orrhage model rats,and to observe whether acupuncture had protection on brain tissues. Methods: 84 male SD rats were randomly divided into 5 groups: blank groups,sham operation group,the model group,acupuncture group (model and acupuncture group) and inhibitor group (SP600125). Each group had 24 rats. The mode rats of autologous blood cerebral hemorrhage were made, and after modeling success,they were acupunctured through the three parts of frontal,parietal and temporal with one needle by the method of acupuncture penetrating Baihui (GV20) and Qubin(GB7). After successful modeling, acupuncture was taken at four points: 6 h, 3 d and 7 d, and the gray value of p-STAT3 positive cells in brain tissue was observed by immunohistochemistry (IHC). Results: Acupuncture penetrating Baihui (GV20) and Qubin (GB7) could reduce the nerve defect symptoms and improve the score of model animals. The expression of p-STAT3 began to increase in the six hours after intracerebral hemorrhage in rats, and the expression was the most in the first day, and decreased rapidly after reaching the peak on the third day, and still expressed on the seventh day. The gray value of the positive cells in the acupuncture group was lower than that in the model group at each time point, and the difference was significant. There was statistical significance between acupuncture group and SP600125 group. Conclusion: Acupuncture penetrating Baihui(GV20) and Qubin(GB7) can reduce the degree of neurologic impairment and reduce the expression of p- STAT3 protein in rats,which indicates that acupuncture plays a neuroprotective role in the treatment of intracere- bral hemorrhage.
