百合疏肝安神汤对焦虑性抑郁症模型大鼠细胞因子及单胺类神经递质表达的影响 被引量:17

Effects of Compound Baihe Shugan Anshen on the expression of cytokines and monoamine neurotransmitters in anxious depression rats model

摘要 目的研究百合疏肝安神汤对焦虑性抑郁症模型大鼠细胞因子及单胺类神经递质表达的影响。方法 SD大鼠按体重被随机分为7组:模型组、阳性药对照组(文拉法辛6.75 mg·kg^(-1))、高中低3个剂量实验组(百合疏肝安神汤,24.28,12.24,6.12 g·kg^(-1)),空白对照组(正常饲养)和溶媒组(皮下注射0.9%NaCl+5%二甲基亚砜)。经连续21 d慢性束缚应激+孤笼饲养,并皮下注射皮质酮(30 mg·kg^(-1))来复制焦虑性抑郁症动物模型。造模同时,灌胃给药。用酶联免疫吸附法检测血浆、前额叶皮层(PFC)、海马和杏仁核组织中肿瘤坏死因子-α(TNF-α)含量;以免疫印迹法检测PFC、海马和杏仁核组织中吲哚胺2,3-双加氧酶(IDO)表达;用HPLC-ECD法测定前额叶皮层、海马和杏仁核中单胺类神经递质5-羟色胺(5-HT)、多巴胺(DA)和去甲肾上腺素(NE)的含量。结果血浆TNF-α含量、PFC中IDO的OD值、PFC中5-HT含量、海马中DA含量、杏仁核中NE含量:空白对照组与模型组分别(53.00±7.36),(96.75±10.10)pg·mL^(-1);0.40±0.06,0.79±0.08;(477.10±63.56),(231.94±31.51)ng·mg^(-1);(432.08±30.00),(249.41±25.35)ng·mg^(-1);(829.55±116.67)ng·mg^(-1),(461.49±70.00)ng·mg^(-1),与空白对照组比较,模型组差异均有统计学意义(P<0.05或P<0.01)。血浆TNF-α含量、PFC的IDO的OD值、PFC的5-HT含量、海马DA含量、杏仁核NE含量:高剂量实验组分别为(73.62±7.46)pg·mL^(-1)与(0.55±0.08)及(362.60±42.53),(328.85±30.22),(664.40±65.05)ng·mg^(-1),与模型组比较差异均有统计学意义(P<0.05或P<0.01)。结论百合疏肝安神汤可能是通过调节大鼠血浆和大脑组织中细胞因子和单胺类神经递质,起到抗焦虑抑郁的作用。 Objective To investigate the effects of Compound Baihe Shugan Anshen( CBSA) on the expression of cytokines and monoamine neurotransmitters in anxious depression model rats. Methods According to weight,SD rats were randomly divided into 7 groups: model group,positive control group( venlafaxine,6. 75 mg·kg^-1),experimental-H,M,L groups( CBSA,24. 28,12. 24,6. 12 g·kg^-1). Chronic restraint stress + solitary feeding for 21 days, and subcutaneous injection corticosterone 30 mg·kg^-1 were used to replicate the anxiety depression animal model. Intragastric administration in the modeling time animals were treat as ig 0. 9% normal saline + 5% dimethyl sulfoxide( DMSO)in the vehicle group,and normal feeding in the control group. The tumor necrosis factor-α( TNF-α) content of plasma,prefrontal cortex( PFC),hippocampus and amygdala was tested by ELISA method. The expression of idoleamine 2,3-dioxygenase( IDO) in amygdala and hippocampus tissues was assayed by Western-blotting. The content of 5-hydroxytryptamine( 5-HT),dopamine( DA) and norepinephrine( NE) of PFC,hippocampus and amygdala were analysed by HPLC-ECD. Results The TNF-α content of plasma,IDO optical density of PFC,5-HT content of PFC,DA content of hippocampus and NE content of amygdala in blank control group and model group respectively were( 53. 00 ± 7. 36),( 96. 75 ± 10. 10) pg · mL^-1; 0. 40 ± 0. 06, 0. 79 ± 0. 08;( 477. 10 ± 63. 56),( 231. 94 ± 31. 51) ng · mg^-1;( 432. 08 ± 30. 00),( 249. 41 ± 25. 35) ng · mg^-1;( 829. 55 ± 116. 67),( 461. 49 ± 70. 00) ng·mg^-1. Compared with blank control group,the TNF-α content of plasma,PFC,hippocampus and amygdala were increased significantly; the IDO optical density of PFC,hippocampus and amygdala were increased significantly; the monoamine neurotransmitters of PFC,hippocampus and amygdala were reduced significantly in model group,the difference between the two groups was statistically significant( P〈0. 05 or P〈0. 01). The above five factors in experimental-H group respectively were( 73. 62 ± 7. 46) pg · mL^-1,0. 55 ± 0. 08 and( 362. 60 ± 42. 53),( 328. 85 ± 30. 22),( 664. 40 ± 65. 05) ng ·mg^-1. Compared with the model group,hippocampus and amygdala were increased in the experimental-H group, but another three factors in experimental-H group significantly were reduced,the difference between the two groups was statistically significant( P〈0. 05 or P〈0. 01). Conclusion CBSA regulates plasma and brain tissue cytokines and monoamine neurotransmitters,which may play a role in antianxiety and depression.
