摘要
目的建立化妆品中米诺地尔的毛细管区带电泳(CZE)测定法。方法样品经提取液(甲醇∶2 mmol/L磷酸二氢钠+3 mmol/L磷酸体积比为1∶9)提取后离心,过0.45μm膜后上机。采用未涂敷熔融石英毛细管[75μm×30.2 cm(有效长度:20 cm)];分离缓冲液为20 mmol/L磷酸二氢钠-30 mmol/L磷酸,pH 2.13;分离电压为11 kV;检测波长为278 nm;进样压力为0.5 psi;进样时间为3 s。结果在1~160 mg/L的线性范围内,所得米诺地尔的回归方程为y=151.12x+73.363,r=0.999 95,呈良好的线性关系。该方法的检出限为0.3 mg/L,定量下限为1.0 mg/L,平均回收率在93.4%~95.9%之间,RSD在3.3%~3.7%之间。结论该方法简单、快速、准确,可用于化妆品中米诺地尔含量的检测。
Objective To establish a method for the determination of minoxidil in cosmetics by capillary zone electrophoresis. Methods Samples were extracted by a solution of V (methanol) :V (2 mmol/L NaH2PO4+3 mmol/L H3PO4)=1:9 and then centrifuged. The supernatant was filtered through 0.45μm membrane and then directly injected. The separation was carried out on an uncoated fused-silica capillary [75 μm (i.d.) 30.2 cm total length (effective length: 20 cm)]. The separation buffer consisted of 20 mmol/L NaH2PO4 and 30 mmol/L H3PO4, pH 2.13. The separation voltage was 11 kV and the detection wavelength was set at 278 nm. The injection pressure was 0.5 psi and the injection time was 3 s. Results The method had a good linearity and reproducibility in the ranges of 1-160 mg/L and the correlation coefficients (r) were 0.999 95. The limit of detection (LOD, S/N=3) and limit of quantitation (LOQ,S/N=10) were 0.3 mg/L and 1 mg/L, respectively. The average recovery were between 93.4% and 95.9% with relative standard deviations (RSDs) ranging from 3.3% to 3.7%. Conclusion The method is suitable for the rapid and accurate determination of minoxidil in cosmetics with simple sample pretreatment.
