摘要 目的探讨星形细胞上调基因-1(AEG-1)与P53、Ki67在胶质瘤中的表达,及其表达的相关性和临床意义。方法采用免疫组化检测39例胶质瘤组织和10例正常脑组织AEG-1、P53、Ki67蛋白的表达,分析AEG-1、P53、Ki67表达与胶质瘤病理级别的关系。PCR检测AEG-1 mRNA在不同级别胶质瘤组织的表达水平。结果 AEG-1、P53、Ki67在正常脑组织中均未检测到,在胶质瘤组织的表达率随胶质瘤级别的增高而增高(P<0.001);AEG-1阳性组P53、Ki67的表达高于AEG-1阴性组,AEG-1的表达与P53、Ki67表达呈正相关(r=0.349,P<0.05;r=0.659,P<0.001)。AEG-1 mRNA在高级别胶质瘤组织的表达水平明显高于低级别胶质瘤(均P<0.05)。结论 AEG-1与P53、Ki67在高级别胶质瘤组织中的表达率增高,AEG-1与P53、Ki67的表达呈正相关,可作为肿瘤发生、发展过程中较敏感的生物学指标。 Objective:To analyze expression and clinical significance of AEG-1 correlating with P53 and Ki67 in glioma. Methods: The expression of AEG-1,P53 and Ki67 protein in 39 cases of human glioma tissues and 10 cases of normal brain tissues by immunohistochemistry were detected.The expression of AEG-1 had correlation with P53,Ki67 and the clinicopathologic features of glioma were analyzed.The expression levels of AEG-1 mRNA in different grades of glioma tissues by real-time fluorescence quantitative PCR were detected. Results:AEG-1,P53 and Ki67 were not detected in normal brain tissues.While in glioma tissues,expression of AEG-1,P53 and Ki67 was strengthened significantly with the improvement of grade of glioma (P〈0.05).In AEG-1 positive group,the expression of P53 and Ki67 was higher than that in AEG-1 negative group,which indicated that AEG-1 expression was positively correlated with P53 and Ki67(P〈0.05).The fluorescence quantitative PCR showed that the expression of AEG-1 mRNA in Ⅲ-Ⅳ glioma tissues were higher than that inⅠ-Ⅱglioma tissues. Conclusion:The expression of AEG-1,P53 and Ki67 in glioma tissues is increased with the improvement of grade of glioma,and AEG-1 is positively correlated with P53 and Ki67 which can be used as a sensitive indicator in tumor development.
