LncRNA MEG3在甲状腺乳头状癌中的表达及对增殖、侵袭与转移能力的影响 被引量:4

摘要 目的探讨lnc RNA MEG3在甲状腺乳头状癌中的表达意义以及对甲状腺乳头状癌细胞增殖、侵袭转移能力能力的影响。方法收集2012年5月~2015年12月于遵义医学院第五附属珠海医院手术切除的甲状腺乳头状癌标本以及对应的癌旁组织,采用real-time PCR技术分别检测68例甲状腺乳头癌组织中MEG3表达情况,并分析其表达情况与临床病理参数的关系;通过细胞转染技术构建MEG3高表达甲状腺癌TPC-1细胞系,利用CCK-8方法检测MEG3对甲状腺乳头状癌细胞增殖能力的影响;采用Transwell方法检测MEG3对甲状腺乳头状癌细胞侵袭迁移与转移能力的影响;利用Western blot方法检测过表达MEG3基因后对甲状腺乳头状癌细胞Snail2蛋白表达的影响。结果 lnc RNA MEG3在甲状腺乳头状癌组织中的表达水平显著低于癌旁组织,差异有统计学意义(P<0.05);MEG3在甲状癌组织中的表达与肿瘤大小、TNM分期有关(P<0.05),与其他病理参数无关(P>0.05);TPC-1细胞系中过表达MEG3可使甲状腺乳头状癌细胞的增殖能力降低,并且能够抑制甲状腺乳头状癌细胞的侵袭与转移能力(P<0.05)。real-time PCR和Western blot检测发现,过表达MEG3能够显著抑制Snail2基因与蛋白的表达(P<0.05)。结论 lnc RNA MEG3在甲状腺乳头状癌中表达显著降低且与甲状腺的发生发展相关。MEG3能够显著抑制甲状腺乳头状癌细胞的增殖能力,且能够通过抑制Snail2蛋白的表达抑制状腺癌细胞的侵袭与转移能力。 Objective To investigate the expression of lncRNA MEG3 in papillary thyroid carcinoma and explore the effect of MEG3 on proliferation, migration and invasion in PTC-1 cells. Methods 68 cases of papillary thyroid carcinoma specimens and paired normal tissue were collected after surgery from May 2012 to December 2015 in the Fifth Affiliated Hospital of Zunyi Medical College. The expression of MEG3 was analyzed by quantitative real-time PCR. The effect of MEG3 on proliferation in TPC-1 cells was detected by CCK-8 method. The effect of MEG3 on migration and invasion abilities of TPC-1 cells were analyzed by Transwell assays. The expression of Snail2 was detected by Western blot. Results The expression of MEG3 was lower in papillary thyroid carcinoma tissues than adjacent normal tissues (P 〈 0.05). The expression of MEG3 was related with TNM stage and tumor size (P 〈 0.05). Over expressed MEG3 significantly inhibited the proliferation of TPC-I ceils (P 〈 0.05). And MEG3 could decrease the abilities of migration and invasion by down regulating the expression of Snail2 (P 〈 0.05). Conclusion The expression of lncRNA MEG3 is lower in the papillary thyroid carcinoma and can inhibit the proliferation, migration and invasion of TPC-1 cells can be inhibited by the expression of Snail2.
