摘要
(1)目的探讨重组人粒细胞集落刺激因子(rhG-CSF)对脑出血大鼠炎症损伤的影响及其机制。(2)方法选择健康雄性大鼠60只,随机分为对照组、模型组和观察组。模型组和观察组采用右侧脑组织基底节内注入新鲜自体不凝血复制脑出血模型,对照组仅经注射点注入生理盐水。脑出血建模成功后观察组在术后6小时开始每天腹部皮下注射rhG-CSF。另两组腹部皮下注射等量生理盐水。采用干湿重法测定各组大鼠病灶周围脑组织含水量(BWC),用免疫组织化学法及免疫蛋白印记法观察大鼠病灶周围脑组织Toll样受体-4(TLR-4)、核因子-κB受体(NF-κB)的免疫阳性细胞数及蛋白表达水平。(3)结果观察组术后各时间点BWC值均低于模型组,差异有统计学意义(P<0.05)。观察组术后病灶周围脑组织TLR-4、NF-κB阳性细胞计数均低于模型组,差异有统计学意义(P<0.05);TLR-4、NF-κB蛋白表达量均明显低于模型组,差异有统计学意义(P<0.05)。(4)结论 rhG-CSF可能通过降低TLR-4、NF-κB的表达,减轻继发性脑水肿及神经细胞损伤,对脑出血大鼠脑组织起保护作用。
Objective To explore the effect and mechanism of recombinant human granulocyte colony stimulating factor(rhG-CSF)on inflammatory injury in rats with intracerebral hemorrhage(ICH).Methods Sixty healthy male rats were randomly divided into control group,model groupand observation group,the model group and observation group were adopted the right brain tissue in the basal ganglia to injected fresh autologous blood to copy the model of intracerebral hemorrhage.The control group were injected with saline.After treatment,the patients in the observation group received abdominal subcutaneous injection of rhG-CSF every day after 6 h.The other two groups were injected with normal saline.Determination of brain tissue around the lesion rats water content by dry wet weight method(BWC),by immune histochemistry and western blot technique,observed the Toll like receptor in rat brain tissue around the lesion in 4(TLR4),the expression level of the positive cell number and protein NF-kappa B.Results Recombinant human granulocyte colony-stimulating factor observation group at each time point after operation BWC values were lower than the model group(P 0.05);the observation group after brain lesions around TLR4,NF-kappa B positive cell counts were lower than the model group(P 0.05),and around the lesion of brain tissue TLR4 and NF-kappa B protein expression was significantly lower than the model group(P 0.05).Conclusion Hecombinant human granulocyte colony-stimulating factor may be through reducing the expression TLR4,NF-kappa B,alleviate cerebral edema and nerve cell injury,protective effect on brain tissue of rats with brain hemorrhage.
