摘要
[目的]探讨氧化槐果碱(OSC)改善角叉菜胶诱导小鼠炎性疼痛的作用机制。[方法]ICR小鼠30只,随机分为阴性对照组、模型组、氧化槐果碱治疗组。采用角叉菜胶诱导的小鼠足肿胀试验,研究氧化槐果碱对小鼠炎性疼痛的影响。[结果]与模型组相比,HE染色结果显示OSC显著抑制损伤足组织的水肿并减少损伤组织中中性粒细胞的数量;Q-PCR结果显示OSC显著降低了COX-2mRNA表达水平(P<0.05);免疫组化分析显示OSC降低了损伤足组织中p-ERK1/2、COX-2的平均积分光密度(P<0.05);WesternBlot分析可知,OSC显著降低损伤足组织中p-ERK1/2、COX-2的蛋白表达量(P<0.01)。[结论]氧化槐果碱对小鼠炎性疼痛具有明显改善作用,可能与调节ERK1/2-COX-2-PGE_2通路有关。
[Objective] To uncover the underling mechanism of ameliorative effects of oxysophocarpine(OSC) on carrageenaninduced inflammatory pain in mice. [Methods]A total of thirty ICR mice were randomly allocated into the following three groups:the control group(n=10), the inflammatory pain model group(n=10), and the OSC intervention group(n=10). The carrageenaninduced paw edema model was used to investigate the effects of OSC on inflammatory pain in mice. [Results] HE staining demonstrated that in OSC intervention group, the paw edema was significantly suppressed and the numbers of neutrophils in injured tissues were decreased; real-time quantitative PCR assay revealed that the m RNA expression level of COX-2 was significantly inhibited by OSC(P <0.05); the immunohistochemical assay showed that OSC significantly suppressed the mean integral optical density of p-ERK1/2 and the COX-2 in injured paw tissues(P<0.05); Western Blot analysis demonstrated that the expression levels of p-ERK1/2 and COX-2 protein were significantly down-regulated in injured paw tissues by OSC(P <0.01). [Conclusion] The combined data suggest that OSC attenuates inflammatory pain by adjusting the ERK1/2-COX-2-PGE_2 signaling pathway in mice.
