摘要
目的:探讨结肠癌组织中双糖链蛋白聚糖(Biglycan)的表达与上皮间质转化(EMT)的关系。方法:收集8对结肠癌同患者的癌及癌旁组织,采用免疫组织化学、实时荧光定量聚合酶链式反应(Real-time PCR)和蛋白质免疫印迹(Western blot)检测组织中Biglycan、上皮标志物E-钙黏素(E-cadherin)、间质标记物N-钙黏素(N-cadherin)和波形蛋白(vimentin)的表达,分析结肠癌组织中Biglycan与细胞EMT的相关性。结果:与癌旁组织相比,结肠癌组织中Biglycan的表达显著上调(P<0.05);E-cadherin的表达显著降低(P<0.05);间质标记物N-cadherin、vimentin表达显著升高(P<0.05)。结论:Biglycan在结肠癌组织的表达上调可能是通过启动EMT的发生来实现,并促进肿瘤的侵袭转移。
Objective: To investigate the relationship between the Biglycan expression of colon cancer and transformation of epithelial stromal. Methods: Colon cancer and peficancerous tissues of 8 patients were collected to test Biglycan in tissue,epithelial markers E-cadherin,interstitial markers N-cadherin as well as the expression of vimentin and analyze the correlation between Biglycan in colon cancer tissue and epithelial mesenchymal transition by immunohistochemistry,real-time PCR and Western blot.Results: Compared with adjacent tissues,the expression of biglycan was significantly upregulated in colon cancer tissues( P〈0. 05),and the expression of E-cadherin was significantly decreased( P〈0. 05). The expression of N-cadherin and vimentin were significantly increased( P〈0. 05). Conclusion: The up-regulation of Biglycan expression in colon cancer may be triggered by the initiation of EMT and promotes tumor invasion and metastasis.
