摘要
目的研究青藤碱对骨质疏松压缩性骨折模型家兔骨生长分化因子2(GDF2)和血清白介素-1β(IL-1β)及IL-17的影响,并探讨其作用机制。方法先将36只雌性家兔去卵巢建立骨质疏松模型,4周后再采用剪断L1-L5节段椎体前缘的方法建立骨质疏松压缩性骨折模型,成模后分为模型组,青藤碱A组、B组。青藤碱A组、B组用10%青藤碱分别按2、4 m L/(kg·d)剂量灌胃,模型组灌胃等体积生理盐水,三组均治疗1个月。采用双抗体夹心法检测耳缘静脉炎性因子IL-1β/IL-17的含量;采用酶联免疫吸附(ELISA)法检测椎间盘组织GDF2表达水平;采用逆转录-聚合酶链式反应(RT-PCR)法检测椎间盘组织GDF2 m RNA表达。结果青藤碱可提高青藤碱A组、B组家兔GDF2、GDF2 m RNA表达水平,降低血清IL-1β/IL-17含量,与模型组和同组治疗前比较,差异有统计学意义(P<0.05),但青藤碱A组、B组比较差异无统计学意义(P>0.05)。结论青藤碱可改善低IL-1β/IL-17介导的炎性反应及骨退行性变,提高GDF2诱导成骨分化和骨形成能力,加速骨折愈合。
Objective To study the effects of sinomenine on growth differentiation factor 2( GDF2) and IL-1β and IL-17 in models of osteoporotic vertebral compression fractures in rabbits,and to explore the mechanism of action. Methods First,36 female rabbits were ovariectomized to establish osteoporosis models,and after 4 weeks,the method of cutting former-edge of L1-L5 segments was used to establish fractures of vertebral compression with osteoporosis. After modeling,they were divided into model group,sinomenine group A and group B. Sinomenine group A and group B received intragastric administration of 10% sinomenine with 2 m L/( kg·d) and 4 m L/( kg·d) as dosages,model group was given normal saline,and the 3 groups were all treated for one month. Double antibody sandwich method was used to test the contents of factor IL-1β/IL-17 of ear vein inflammatory; enzyme linked immunosorbent assay( ELISA) was used for the detection of expressions of GDF2 in intervertebral disc tissue; reverse transcription polymerase chain reaction( RT-PCR) was used to detect the expressions of GDF2 m RNA in intervertebral disc tissue. Results Sinomenine could improve the expression level of GDF2 and GDF2 m RNA in group A and group B,and decrease the contents of IL-1β/IL-17 in serum( P〈0. 05),and there was statistically significant difference between sinomenine groups and model group( P〈0. 05),but there was no significant difference between group A and group B( P〉0. 05). Conclusion Sinomenine can reduce the inflammatory reaction and bone degeneration induced by factor IL-1β/IL-17,increase the ability of GDF2 to induce osteogenic differentiation and bone formation,thus accelerating the healing of bone fractures.
