摘要
目的 :代谢综合征(MS)痰证造模方法的探究,分析高糖高脂喂养及高脂乳剂灌胃后对MS痰证造模的影响。方法 :70只清洁级wistar雄性大鼠在测完体重,空腹血糖(FBG),腹围,血清TG、HDL-C后,随机分为正常组(NS组)10只,模型组(MS组)60只。从第5周开始每2周监测体重、空腹血糖(FBG)、腹围,禁食12 h后眼睑采血测血清TG、HDL-C。高脂高糖饮食喂养13周禁食12 h后眼睑采血测血清胰岛素(INS)并计算HOMA-IR。因饮食喂养后MS组大鼠TG及FBG无升高,15周开始予高脂乳剂灌胃2周,检测体重、腹围、FBG值,禁食12 h后眼睑采血测血清TG、HDL-C、INS,计算HOMA-IR,并分析灌高脂乳剂干预前后各指标变化。结果 :13周后MS组与NS组比较:HDL-C显著降低,体重、腹围均值升高无显著差异,INS、HOMA-IR值显著升高(P<0.01)。高脂乳剂干预后MS组与NS组大鼠各指标比较,腹围显著增大(P<0.05),体重减轻,TG、HDL-C值均显著减小,INS、HOMAIR值显著升高(P<0.01),最终造模成功40只。结论 :高脂乳剂灌胃后未能促进大鼠TG及FBG的升高,高糖高脂饲料喂养是较为稳定、可靠、操作易行的代谢综合征造模法。
Objective :It is to explore the modeling method of metabolic syndrome(MS)phlegm syndrome and analyze the influence high glucose and high fat feeding and high-fat emulsion gavage in MS phlegm syndrome modeling. Methods :60 clean-level wistar male rats are randomly divided into a normal group of 10 rats(NS group)and a model group of 60 rats(MS group)after measuring body weight,FBG and abdominal girth,and sampling eyelid blood and serum TG and HDL-C after abrosia for 12 h. The body weight,FBG,abdominal girth,and eyelid serum TG and HDL-C were measured and monitored after 12 h abrosia every two weeks from the 5 th week. After high glucose and high fat feeding for 13 weeks,eyelid serum samples after 12 h abrosia were used to measure serum INS and calculate HOMA-IR. Because the TG and FBG of rats in MS group have no increase after diet feeding,then from 15 th week,2 weeks high fat emulsion gavage is applied to observe biochemical indicators before and after intervention. Results :After 13 weeks,compared with NS group,MS group has an obvious decrease in HDL-C,an increase in mean value of body weight and abdominal girth but without obvious difference,an obvious increase in INS and HOMA-IR values(P0.01). After the intervention of high fat emulsion,compared with various indexes of the rats in NS group,MS group has obvious increases in abdominal girth(P0.05)and in INS and HOMA-IR values(P0.01),obvious decreases in body weight andin TG and HDL-C values,respectively. Finally :40 metabolic syndrome phlegm syndrome rats are successfully modeled. Conclusion :The high fat emulsion gavage fails to promote the rats' increase in TG and FBG,high glucose and high fat diet feeding is a relatively stable,reliable and operable modeling method of rat metabolic syndrome phlegm syndrome.
