摘要
目的观察低温复合丙泊酚对大鼠海马神经元凋亡蛋白Caspase-3、自噬相关蛋白Beclin 1和LC3-Ⅱ及其神经元超微结构变化的影响,探讨丙泊酚对低温所致大鼠海马神经元自噬与凋亡的干预作用。方法将大鼠分为空白对照组(A组)、低温丙泊酚组(B组)和低温水合氯醛组(C组),B、C两组低温干预30min,各组进行心脏灌注,断头取脑,制备大鼠海马神经元组织标本,用蛋白免疫印迹法(Western blot)检测Caspase-3的表达,用Western blot检测Beclin-1和LC3-Ⅱ蛋白的表达,透射电镜观察神经元超微结构的变化。结果 3组Caspase-3、Beclin-1和LC3-Ⅱ蛋白的水平差异有统计学意义(P<0.05);透射电镜结果显示B、C组大鼠海马区神经元均有不同程度的改变,尤其是C组神经细胞凋亡现象明显。结论低温状态下海马神经元仍有一定的自噬与凋亡现象,而丙泊酚则可减少这种损害,具有较好的神经元保护作用。
Objective To observe the effect of low temperature compound propofol on the changes of apoptosis protein Caspase-3,autophagy-related proteins Beclinq and LC3-Ⅱ and thier changes of hippocampal neurons. Methods The rats were randomly divided into blank control group (Group A), propofol group at low temperature (Group B) and chloral hydrate group at low temperature (Group C),Group B and C were treated with low temperature for 30 rain. Then, each group was subjected to cardiac perfusion and decapitated brain to prepare rat hippocampal neuronal tissue samples. The expression of Caspase-3 was detected by immunohistochemistry,Beclin-1 and LC3-Ⅱ protein was detected by immunoblotting. The ultrastructural changes of neurons were observed by transmission electron microscopy. Results The expression of Caspase-3, Beclin-1 and LC3-Ⅱ protein in each group were statistically significant differences (P〈0.05). The results of transmission electron microscopy showed that the neurons in group β and C were changed in different degrees,especially in group C neuronal apoptosis is obvious. Conclusion Autophagy and apoptosis in existence still exist in low temperature condition, while propofol can reduce this damage and have better protective effect on neurons.
