摘要
目的 探讨创伤性急性肺损伤 (ALI)肺泡巨噬细胞 (PAM)核因子 (NF) κB的活化和细胞因子的释放及地塞米松 (Dex)的抑制作用。方法 将 2 4只大耳白兔随机分为正常对照组、创伤组、Dex干预组。用凝胶电泳迁移率改变分析法 (EMSA)和酶联免疫吸附 (ELISA)法分别检测PAM核提取物中NF κB的活性和PAM培养上清液中肿瘤坏死因子 (TNF) α、白细胞介素 (IL) 6、IL 1 0的含量。结果 NF κB活性和TNF α、IL 6、IL 1 0的含量创伤组分别为 (2 987.85± 1 63 .85)ng/L、(2 35 .6± 30 .8)ng/L、(398.8± 2 4 3 .6)ng/L、(2 4 6 .4± 30 .5)ng/L ,较对照组比较均显著增高(P <0 .0 1或P <0 .0 5) ;Dex组NF κB(1 968.2 7± 69.42 )ng/L、TNF α为 (1 68.2± 48.8)ng/L、IL 6为 (2 1 0 .3± 2 8.6)ng/L较创伤组比较明显降低 (P <0 .0 1 ) ,但IL 1 0 (2 2 7.2± 38.6)ng/L无明显变化 (P >0 .0 5)。结论 NF κB激活和分泌高水平细胞因子在创伤性ALI的发生、发展过程中起着重要作用 ;Dex发挥抗炎作用与其对NF
Objective To investigate the activation of nuclear factor κB (NF κB) and the changes of cytokine levels in pulmonary alveolar macrophages (PAM) in traumatic acute lung injury (ALI) and the effect of dexamethasone (Dex).Methods Twenty four rabbits were randomly divided into the normal group, traumatic ALI group, Dex treatiment group. The NF κB activity of nuclear protein extracted from the PAM and the concentration of tumor necrosis factor α (TNF α), interleukin 6 (IL 6), IL 10 in the supernatant were measured by electrophoretic mobility shift assay (EMSA) and enzyme linked immune adsorbing analysis (ELISA),respectively. Results The activity of NF κB and the levels of TNF α, IL 6 and IL 10 [(2?987.85±163.85)?ng/L,(235.6±30.8)?ng/L,(398.8±243.6)?ng/L,(246.4±30.5)?ng/L,respectively] in traumatic ALI group were significantly higher than those in normal group [(922.02±28.36)?ng/L,(36.4±8.0)?ng/L,(78.6±39.4)?ng/L,(190.3±25.6)?ng/L,P<0.01 or 0.05].In Dex treatiment group, the activity of NF κB and the levels of TNF α, IL 6 [(1?968.27±69.42)?ng/L,(168.2±48.8)?ng/L,(210.3±28.6)?ng/L respectively] were significantly lower than those in traumatic ALI group (P<0.01), but no significant change was found in IL 10 content (P>0.05).Conclusion NF κB activation and cytokine secretion of alveolar macrophages may play an important role in development and progression of traumatic ALI; Dex shows anti inflammatory effect in traumatic ALI, which is closely ralated to the regulatory role of Dex on the NF κB activation and cytokine secretion.
