摘要
目的构建核桃楸皮的HPLC指纹图谱并研究不同溶剂与煎煮方式对满药木鸡汤(核桃楸皮-云芝)药对活性组分溶出的影响。方法利用核桃楸皮的3种有效组分胡桃醌、槲皮素、山奈酚的标准品构建HPLC指纹图谱,分别以乙酸乙酯、甲醇、水为提取溶剂进行活性组分提取,研究单煎与合煎2种提取方式所得提取物的组分差异。结果乙酸乙酯、甲醇、水3种提取溶剂的核桃楸皮单煎提取率分别为8.17%、18.51%、14.77%,均高于药对的合煎提取率3.89%、10.90%、14.24%,合煎提取会影响核桃楸皮活性组分的溶出。基于HPLC指纹图谱确定了核桃楸皮中胡桃醌、槲皮素、山奈酚3种功能组分的单煎得率与合煎得率比值,在甲醇提取中比值为2.3、2.2、2.9,在乙酸乙酯提取中分别为3.2、2.5、2.5,在水提取中分别为4.9、2.3、1.4,与药对中的云芝合煎提取会影响核桃楸皮功能组分的溶出。基于HPLC指纹图谱发现,在3种溶剂提取结果中,药对合煎提取均会造成核桃楸皮中的部分功能组分不能有效溶出,同时也在合煎提取物中检测到新组分。结论木鸡汤(核桃楸皮-云芝)药对的合煎提取会降低核桃楸皮功能组分的浸出,分煎提取后将活性组分合并使用可提高核桃楸皮功能组分的使用效率。
Objective The HPLC fingerprints of extracts of the barks of Juglans mandshurica Maxim. were constructed and used to study the effects of different solvents and methods of decocting on the dissolution of active components of the materials of Manchurian Muji Decoction( Juglans mandshurica Maxim.-Coriolus versicolor). Methods Based on the standard materials of the Juglone,Quercetin and Kaempferia,the major three functional components of the barks of Juglans mandshurica Maxim.,the HPLC fingerprints of the crude extracts were constructed. With the ethyl acetate,methanol and water as extraction solvents,the crude extracts were obtained respectively by different decocting methods. Results The experiment results showed that the extraction rates of the barks of Juglans mandshurica Maxim. in ethyl acetate,methanol and water solvents were 8. 17%,18. 51% and 14. 77%,respectively. But,the extraction rates became to 3. 89%,10. 90% and 14. 24% when the bark material were combined with the Coriolus versicolor( another material of Manchurian Muji decoction). When the two materials were mixed,the Coriolus versicolor inhibited the extraction of the functional components of the barks of Juglans mandshurica Maxim. Based on the HPLC fingerprint,the yields of the three functional compounds( Juglone,Quercetin and Kaempferia) of the barks of Juglans mandshurica Maxim extracted without or with Coriolus versicolor were obtained,and the ratio( without/with Coriolus versicolor)were 2. 3,2. 2 and 2. 9 in methanol solvents,3. 2,2. 5 and 2. 5 in ethyl acetate solvents,and 4. 9,2. 3 and 1. 4 in water solvents,respectively. The three active components of Juglans mandshurica Maxim. could not be extracted efficiently when mixed with Coriolus versicolore. Conclusion Based on the HPLC fingerprint of crude extracts,we found that some components of Juglans mandshurica Maxim. could not be obtained when the two materials were combined to extract. But,new components could be detected in crude extracts of the mixed materials of Juglans mandshurica Maxim.-Coriolus versicolor. Conclusion In Manchurian Muji Decoction( Juglans mandshurica Maxim.-Coriolus versicolor),the extraction of the functional components in the barks of Juglans mandshurica Maxim. were inhibited by Coriolus versicolor when the two materials were extracted together. It was better to extract respectively,and then combine the extracts to raise the contents of the active components.
