Study on Synthesis of Targeted Drug Carrier Based on N-trimethyl Carboxymethyl Chitosan Having Pendant Galactose Residues

摘要 以羧甲基壳聚糖为原料,与3-氯-2-羟丙基三甲基氯化铵、乳糖反应,用KBH4还原,合成半乳糖季铵化羧甲基壳聚糖,并对其进行结构表征、测定其氨基取代度与溶解性能。结果显示,合成产物IR中有出现三甲基季铵离子中的-CH3吸收峰,NMR中有三甲基季铵离子中的-CH3吸收峰,并有3处出现半乳糖基上的相邻-OH所裂分的双峰,氨基取代度为55.7%,在水中溶解性能较好。结果表明,该方法可合成半乳糖季铵化羧甲基壳聚糖。 N-trimethyl carboxymethyl chitosan was produced by carboxymethyl chitosan and 3-chloro-2-hydroxypropyltrimethylammonium chloride,then it reacted with lactose in its NH2 and KBH4,galactosylated N-trimethyl carboxymethyl chitosan was produced,the chemical structures of galactosylated N-trimethyl carboxymethyl chitosan were characterized by FTIR and1 H-NMR, result proved the synthesis was successful. Galactosylated N-trimethyl carboxymethyl chitosan may be used as drug delivery carriers for hepatocyte-targeting.
