从肠道菌群多样性探讨越鞠丸对ApoE^(-/-)小鼠血脂的影响 被引量:18

摘要 目的:观察治疗六郁的代表方越鞠丸对血脂的影响,为临床应用和实验研究提供参考依据。方法:采用高脂饲料饲养ApoE^(-/-)小鼠的方法建立动脉粥样硬化斑块模型,用越鞠丸和辛伐他汀分别对高脂饲料饲养的ApoE^(-/-)小鼠进行干预,ELISA法检测小鼠血清血脂含量,基于16S rDNA基因测序技术分析小鼠肠道菌群。结果:越鞠丸组、辛伐他汀组和模型对照组小鼠血清胆固醇(TC)、甘油三酯(TG)和低密度脂蛋白(LDL-C)含量明显高于正常对照组,高密度脂蛋白(HDL-C)含量明显低于正常对照组;越鞠丸组小鼠TC和LDL-C含量明显低于模型对照组且与辛伐他汀组比较差异无统计学意义,TG和HDL-C含量与模型对照组比较差异无统计学意义。越鞠丸组与正常对照组、模型对照组、辛伐他汀组小鼠粪便样本菌群存在显著性差异,模型对照组与辛伐他汀组小鼠粪便样本菌群比较差异无统计学意义。结论:越鞠丸能降低TC和LDL-C,其机制与辛伐他汀不同,越鞠丸可能通过抑制红蝽菌科等4类细菌和促进双歧杆菌科等5类细菌的生长来降低TC和LDL-C。 Objective: To observe the effect on blood lipids of Yueju pills, a representative formula of the SixYu, thus provide reference for clinical application and experimental study. Methods: The model of atherosclerotie plaque was established by feeding ApoE-/- mice with high-fat diet. Interventions include Yueju pills and simvastatin. Elisa was used to detect the serum lipid content in mice. The intestinal microflora was analyzed based on 16S rDNA gene sequencing. Results: Miee in Yueju pills group, simvastatin group as well as the model control group experienced higher level of serum cholesterol (TC) , triglyeeride (TG) , low density lipoprotein (LDL-C) and lower level of high-density lipoprotein ( HDL- C) than those in the normal diet control group. TC and LDL-C levels of mice in Yueju pills group were lower than those in model control group but showed no significant difference compared with simvastatin group. In terms of triglyceride (TG) and high-density lipoprotein (HDL-C) , Yueju pills group showed no statistically difference compared with model control group. There was a significant difference in the stool samples between Yueju pills group and the other groups, while the difference between the model control group and the simvastatin group was not significant. Conclusion: Yueju pills can reduce TC and LDL-C, whose mechanism is different from simvastatin. Yueju pills may reduce TC and LDL-C by inhibiting the growth of four kinds of bacteria such as Coriobacteriaceae, while promoting the growth of Bifidobaeteriaceae and promoting growth of five other kinds of bacteria such as bifidobacterium.
