伊犁马GHR基因多态性及其与泌乳性能的关联分析 被引量:3

摘要 马奶具有较高的营养价值,但目前关于马泌乳性能方面的研究很少。本实验对伊犁马生长激素受体(GHR)基因外显子和启动子设计13对引物,以DNA混合池为模板,采用PCR-RFLP法检测188匹伊犁马GHR基因多态基因型,并分析有产奶量记录的73匹伊犁马GHR基因多态性与泌乳性状之间的关系。结果表明:在伊犁马中检测到GHR基因启动子T-562C突变,CC、TC、TT 3种基因型,其基因型频率分别0.309、0.676、0.016;C为优势等位基因,频率为0.646,T基因频率为0.354;虽然各基因型间产奶量并没有显著差异(P>0.05),但各基因型的产奶量依次为TT>CC>TC,基因C突变为T可使产奶量有上升趋势,研究结果可为我国乳用马的选育提供一定的参考。 Mare's milk has a high nutritional value, but there are few research reports on lactation performance of horse until now. This trial was conducted to study the polymorphism of growth hormone receptor (GHR) gene in Yili horse and its relationship with lactation performance. 13 pairs of primers were designed for the exons and promoter of GHR gene of Yili horse. DNA pooling was used as the template, and PCR product was directly sequenced. PCR-RFLP method was used to genotype the polymorphic sites in 188 Yili horses, and then the relationship between genotypes and lactation traits based on 73 of them was analyzed. The result showed that a T-562C mutation in GHR gene promoter was detected in Yili horse. The frequencies of genotype CC, TC and TT were respectively 0.309, 0.66 and 0.016, and C allele was dominant and its frequency was 0.646. The frequency of T gene was 0.354. The results showed that although the milk yield of each genotype was not Significantly different (P〉0.05), it was found that there seems to be a trend in the milk production of each genotype (TT〉 CC〉 CT). The mutation (C〉T) could increase the milk yield. The results can provide an identifiable ground for the Chinese dairy horse breeding efficiently.
