摘要
目的探讨瑞舒伐他汀对H型高血压大鼠血管内皮生长因子(vascular endothelial growth factor,VEGF)的影响。方法选择雄性SD大鼠24只,按照随机数字表法分为对照组、H型高血压组、联合组(H型高血压+瑞舒伐他汀干预),每组8只。H型高血压造模采用自发性高血压大鼠喂食添加2%蛋氨酸的基础鼠粮。应用酶联免疫吸附法测定大鼠血清VEGF水平;免疫组织化学法测定大鼠VEGF表达。结果各组大鼠血清TC、TG、HDL、LDL、同型半胱氨酸水平比较,差异无统计学意义(P>0.05)。联合组血清VEGF水平较对照组和H型高血压组明显升高[(217.50±30.40)ng/L vs(180.93±15.75)ng/L,(187.19±10.93)ng/L,P<0.05)]。各组大鼠VEGF表达均为强阳性,其中联合组VEGF表达强度高于对照组及H型高血压组。结论瑞舒伐他汀可能通过影响VEGF表达来影响H型高血压大鼠血管内皮功能。
Objective To study the effect of rosuvastatin on expression of vascular endothelial growth factor(VEGF)in H type hypertension rats.Methods Twenty-four male SD rats were randomly divided into control group,H type hypertension group and H type hypertension+rosuvastatin group(8 in each group).H type hypertension model was established by feeding rats with rat chow plus 2% methionine.Serum VEGF level was measured by enzyme-link immunosorbent assay and expression of VEGF was detected with immunohistochemial staining.Results No significant difference was found in serum levels of TC,TG,HDL,LDL,Hcy between the control group,H type hypertension group and H type hypertension+rosuvastatin group(P〈0.05).The serum VEGF level was significantly higher in H type hypertension+rosuvastatin group than in control group and H type hypertension group(217.50±30.40 ng/L vs 180.93±15.75 ng/L,187.19±10.93 ng/L,P〈0.05).The expression level of VEGF was significantly higher in H type hypertension+rosuvastatin group than in control group and H type hypertension group.Conclusion Rosuvastatin affects the vascular endothelial function in H type hypertension rats by regulating the expression of VEGF.
