雷公藤甲素对乳腺癌MCF-7细胞增殖和凋亡的影响及其机制 被引量:11

摘要 目的观察雷公藤甲素对人乳腺癌MCF-7细胞增殖和凋亡的影响,并探讨其分子机制。方法 MCF-7细胞经不同浓度雷公藤甲素干预后进行实验,CCK-8法检测增殖抑制率,光学倒置显微镜观察细胞形态的变化,流式细胞术检测细胞凋亡率,q RT-PCR及Western blot检测Bcl-2、Bax、Survivin和Cas-pase-3表达。结果雷公藤甲素在一定范围内以剂量和时间依赖的方式抑制MCF-7细胞增殖;与对照组比较,雷公藤甲素作用后细胞出现凋亡形态学改变;细胞凋亡率显著增高;Bcl-2和Survivin表达明显降低;Bax和Caspase-3表达显著增加。结论雷公藤甲素可抑制MCF-7细胞增殖并诱导凋亡,其机制可能与下调Bcl-2和Survivin表达,上调Bax和Caspase-3表达有关。 Objective To explore the effects and mechanism of triptolide on proliferation and apoptosis of breast cancer MCF-7 ceils. Methods MCF-7 ceils were treated by different concentrations of triptolide. CCK-8 as- say was employed to detect the cell proliferation. The morphological changes were observed by an inverted micro- scope. The apoptosis rate was detected by flow cytometry. Expressions of Bcl-2, Bax, Survivin and Caspase-3 were measured by qRT-PCR and Western blot. Results Triptolide inhibited the proliferation of MCF-7 cells in a dose and time-dependent manner at a suitable range. Triptolide induced morphological changes and apoptosis. Triptolide also down-regulated Bcl-2 and Survivin expressions and up-regulated Bax and Caspase-3 expressions. Conclu- sions Triptolide inhibits proliferation and induces apoptosis of MCF-7 cells, and its mechanism may be related to down-regulation of Bcl-2 and Survivin expressions and up-regulation of Bax and Caspase-3 expressions.
