摘要
目的探讨高浓度葡萄糖诱导的人脐静脉内皮细胞(HUVECs)衰老模型中芳香烃受体(AhR)的变化及其对血管生成相关因子表达的影响。方法采用高浓度葡萄糖溶液(33 mmol/L)处理HUVECs诱导内皮细胞衰老,通过β-半乳糖苷酶染色观察细胞衰老情况,Western blot检测HUVECs中AhR蛋白以及血管生成相关因子低氧诱导因子-1α(HIF-1α)蛋白表达,ELISA检测细胞培养上清液中血管内皮生长因子(VEGF)表达的变化,Matrigel小管生成实验评估小管生成能力,Transwell检测细胞迁移能力。结果与对照组相比,高糖组AhR蛋白表达显著上升,而HIF-1α蛋白表达则显著降低,差异有统计学意义(P<0.05);与对照组相比,高糖组的VEGF蛋白含量表达和NO浓度均显著降低,差异均有统计学意义(P<0.05);高糖组平均小管生成长度和细胞迁移数均较对照组显著减少(P<0.05)。结论在高浓度葡萄糖下培养的HUVECs中AhR蛋白表达明显增加,HIF-1α和VEGF等促血管生成因子明显减少。
Objective To investigate the correlation of blood glucose induced senescence in senescent human umbilical vein endothelial cells(HUVECs)and the expression change of aryl hydrocarbon receptor(AhR)and angiogenesis related factor. Methods Senescence induced by glucose (33 mmol/L) in HUVECs was examined by β-galactosidase staining. Protein expression of AhR and hypoxia-inducible factor-1α(HIF-1α)were analysed by Western blot. The concentrations of vascular endothelial growth factor (VEGF) in culture medium were tested by ELISA. Matrigel test was used to estimate tubule formation. Migration was tested by Transwell test. Results Compared with control group ,the expression of AhR protein of treatment group was significantly increased;the expression of HIF-1α protein of treatment group was significantly decreased;the difference was statistically significant (P<0.05). Compared with control group ,the expression of VEGF protein and the level of NO of treatment group was significantly decreased;the difference was statistically significant (P<0.05);the average length of tubular generated and number of cell migration of treatment group significantly decreased ,the difference was statistically significant (P<0.05). Conclusion The expression of AhR protein in human umbilical vein endothelial cells cultured with high concentration of glucose was significantly increased ,and the angiogenic factors such as HIF-1αand VEGF were significantly decreased.
