摘要
以罗布麻为试材,采用农杆菌介导法将罗布麻eIF-5A基因导入烟草,分别对正常生长条件和NaHCO_3胁迫下转AveIF-5A基因烟草和非转基因烟草的过氧化氢酶(POD)活性、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量进行测定,以期为罗布麻eIF-5A基因功能研究提供参考。结果表明:正常生长条件下,转AveIF-5A基因烟草和非转基因烟草POD活性和MDA含量差异不显著,转基因植株SOD活性显著高于非转基因植株;在NaHCO_3胁迫下,转AveIF-5A基因烟草POD活性和SOD活性均极显著高于非转基因植株,而MDA含量极显著低于非转基因植株。AveIF-5A基因过量表达可以提高转基因烟草的耐盐能力。
To understand the effect of elF-5A gene in response to salt stress, the elF-5A cDNA fragment containing an ORF from Apocynum venetum was transformed into tobacco by Agrobacterium-mediated method. The POD activity, SOD activity and MDA content were measured between transgenic AveIF-5A tobaccos and wild type under normal condition and NaHCO3 stress. The results showed that there was no significant difference in POD activity and MDA content between transgenic and non-transgenic plants under normal condition but SOD activity of transgenic plants increased significantly. Under salt stress, transgenic tobacco over-expressing AveIF-5A gene showed an increase in POD and SOD activity, compared to wild lines. In contrast, transgenic tobacco displayed a decrease in MDA content under NaHCO3 stress. Overall,these results indicated that AveIF-SA gene played a critical role in salt tolerance in trans^enic tobacco.
