摘要
目的:观察青钱柳叶复方对糖尿病大鼠胰岛素抵抗的影响。方法:将60只Wistar大鼠随机分为正常对照组,模型组,青钱柳叶复方低、中、高剂量组,模型组及青钱柳叶复方各剂量组在高热能饲料喂养基础上给予地塞米松腹腔注射造模,青钱柳叶复方低、中、高剂量组分别给予青钱柳叶复方7.8、15.6、31.2 mg/kg·d^(-1)灌胃,42 d后检测血糖、胰岛素、肝糖原、肌糖原并计算胰岛素敏感指数和胰岛素抵抗指数,Tunel检测胰岛β细胞凋亡率。结果:与正常对照组比较,模型组血糖、胰岛素、胰岛素抵抗指数、胰岛β细胞凋亡率明显升高,肝糖原、肌糖原、胰岛素敏感指数明显降低,差异具有显著统计学意义(P<0.01);与模型组比较,低、中、高剂量组血糖、胰岛素、胰岛素抵抗指数、胰岛细胞凋亡率明显降低,肝糖原、肌糖原、胰岛素敏感指数明显升高,中、高剂量组与模型组比较差异具有统计学意义(P<0.05,P<0.01),随剂量增加,作用趋势更明显,但中剂量组和高剂量组比较无显著性差异(P>0.05)。结论:青钱柳叶复方可有效降低2型糖尿病大鼠血糖水平,改善胰岛素抵抗。
Objective:To observe the effects of Qingqian Liuye Prescription on insulin resistance in rats with diabetes mellitus.Methods:60 SPF Wistar rats were randomly divided into the normal control group,model group,Qingqian Liuye Prescription low,middle and high dose groups with each 12 rats,rats in model group and Qingqian Liuye Prescription three dose groups were given dexamethasone 0.8 mg/kg·BW intraperitoneal injection on the basis of high energy feed.The low,middle and high dose Prescription groups were given f Qingqian Liuye7.8 mg//kg·d-1,15.6 mg/kg·d-1,31.2 mg/kg·d^(-1) by gavage for 42 days,then the blood glucose,insulin,liver glycogen,muscle glycogen were determined and insulin sensitivity index and resistance index were calculated,the apoptosis rate of pancreatic islet βcell were determined by Tunel method.Results:Compared with the normal control group,the blood glucose,insulin,insulin resistance index,islet cell apoptosis rate in model group were all increased,insulin sensitivity and liver glycogen,muscle glycogen were all decreased significantly,the differences were statistically significant(P〈0.0);compared with the model group,the blood glucose,insulin,insulin resistance index,islet cell apoptosis rate in three dose of Qingqian Liuye Prescription groups were all decreased,insulin sensitivity index and liver glycogen,muscle glycogen were increased,there were significant difference between the high dose group and middle group on these indexes(P〈0.05,P〈0.0).Conclusion:Qingqian Liuye Prescription can effectively reduce blood glucose in type 2 diabetic rats,improve insulin resistance.
