摘要
目的观察补肝健腰方对腰椎间盘退变大鼠椎间盘基质金属蛋白酶3(MMP-3)、基质金属蛋白酶的组织抑制剂1(TIMP-1)表达的影响。方法 SD大鼠75只,随机分为假手术组、模型组、腰痹通组、强力霉素组、补肝健腰组,每组各15只,成功制备大鼠腰椎间盘退变模型,分别给予腰痹通胶囊、强力霉素、补肝健腰方药物灌胃,检测各组干预前和干预后20、40 d时椎间盘MMP-3、TIMP-1表达。结果干预后20、40 d,腰痹通组、强力霉素组、补肝健腰组椎间盘MMP-3、TIMP-1表达较模型组下降,差异有统计学意义(P<0.05),腰痹通组、强力霉素组、补肝健腰组在干预后20、40 d时椎间盘MMP-3、TIMP-1表达水平,分别与本组干预前比较,差异有统计学意义(P<0.05),补肝健腰组在以上各时间点与腰痹通组、强力霉素组比较,差异无统计学意义(P>0.05)。结论补肝健腰方能降低腰椎间盘退变模型大鼠椎间盘MMP-3、TIMP-1表达,这可能是其防治椎间盘退变疾病的作用机理之一。
Objective To observe the influence of Bugan Jianyao Fang (Liver-tonifying Lumbar- invigorating Decoction) on expressions of intervertebral disc matrix metalloproteinase-3 ( MMP-3 ) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in rats with lumbar disc degeneration. Methods SD rats (n = 75 ) were randomly divided into sham-operation group, model group, Yaobitong (Waist Impedi- ment-Freeing Capsules) group, doxycycline (DOX) group and Bugan Jianyao Fang group (each n = 15). After establishing successfully the model of lumbar disc degeneration in rats, all groups were, respectively, given orally Yaobitong, DOX and Bugan Jianyao Fang. The expressions of intervertebral disc MMP-3 and TIMP-1 were detected in all group after and 20 d and 4 d after intervention. Results After intervention for 20 d and 40 d, the expressions of intervertebral disc MMP-3 and TIMP-1 decreased in Yaobitong group, DOX group and Bugan fianyao Fang group compared with model group (P 〈 0.05 ). The difference in expressions of intervertebral disc MMP-3 and TIMP-1 had statistical significance in Yaobitong group, DOX group and Bugan fianyao Fang group after intervention compared with those in the same groups before intervention ( P 〈 0.05 ). The comparison among Bugan Jianyao Fang group, Yaobitong group and DOX group showed that the difference had no statistical significance ( P 〉 0.05 ) at all time points. Conclusion Bugan Jianyao Fang can reduce the expressions of intervertebral disc MMP-3 and TIMP-1 in rats with lumbar disc degeneration, which may be one of mechanisms of Bugan Jianyao Fang preventing and curing lumbar disc degeneration.
