穿山龙总皂苷对痛风性关节炎大鼠血清生物标志物的影响 被引量:7

Effects of total saponins from rhizoma dioscorea nipponica on biomarkers in serum of gouty arthritis rats

摘要 目的:应用分光光度法和柱前衍生高效液相色谱法,测定穿山龙总皂苷(total saponins from rhizoma dioscorea nipponica,TSRDN)对痛风性关节炎大鼠血清中尿酸、肌酐、乳酸、甘氨酸和色氨酸含量的影响。方法:60只雄性Wistar大鼠随机分为6组,分别为正常组、模型组、TSRDN高剂量组(160 mg·kg^(-1))、中剂量组(80 mg·kg^(-1))、低剂量组(40 mg·kg^(-1)),以及阳性药秋水仙碱组(0.3 mg·kg^(-1))。连续灌胃(ig)给予TSRDN 7 d。d 3 ig给予1 h后,膝关节上方髌上韧带进针,注射0.2 m L浓度为25 mg·m L^(-1)尿酸钠晶体混悬液到关节腔,造成实验性痛风性关节炎模型。甘氨酸和色氨酸的含量测定以2,4-二硝基氟苯(2,4-dinitrofluorebenzene,DNFB)为衍生试剂,采用Diamonsil C18(250 mm×4.6 mm,5μm)色谱柱,以甲醇∶乙腈(4∶6)为流动相A,0.03 mol·L^(-1)醋酸铵缓冲液(p H=6.8)作为流动相B,梯度洗脱,柱温30℃,流速1 m L·min^(-1),检测波长360 nm;尿酸、肌酐和乳酸的含量采用紫外分光光度法检测。结果:在痛风性关节炎模型组,甘氨酸与色氨酸含量显著降低;尿酸、肌酐和乳酸含量显著升高。TSRDN给药组与模型组相比,甘氨酸和色氨酸含量显著升高;尿酸、肌酐和乳酸含量显著降低。结论:穿山龙总皂苷可能通过调控血清中尿酸、肌酐、乳酸、甘氨酸和色氨酸的含量发挥治疗痛风性关节炎的作用。 Objective:To explore the effects of total saponin of rhizoma dioscorea nipponica(RDN) on contents of uric acid,creatinine,lactic acid,glycine and tryptophan in serum of gouty arthritis in rats by high performance liquid chromatography(HPLC) and UV spectrophotometric.Methods:60 Wistar rats were randomly divided into six groups.They were normal group,model group,total saponins groups of high(160 mg·kg^(-1)),middle(80 mg·kg^(-1)) and low(40 mg·kg^(-1)) doses and colchicine group.Total saponins of RDN were given for 7 successive days.An hour after total saponins of RDN were given at the 3 d,0.2 mL 25 mg·mL^(-1) MSU suspension was injected into articular cavity through anadesma of kneecap in the knee-joint to induce model of gouty arthritis.A Diamonsil C18 column(250 mm×4.6 mm,5 μm) was used for the HPLC analysis at 30 ℃,The separation was carried out with the mobile phase consisting of methanol:acetonitrile(4∶6)-ammonium acetate(0.03 mol·L^(-1)) at a flow rate of 1 mL·min^(-1).The eluates were monitored by the wavelength at 360 nm for glycine and tryptophan;The contents of uric acid,creatinine and lactic acid in serum were determined by UV spectrophotometric.Results:In the model group,the levels of glycine and tryptophan were significantly decreased and the contents of uric acid,creatinine and lactic acid were significantly increased.Compared with the model group,glycine and tryptophanincreased significantly while uric acid,creatinine and lactic acid decreased significantly in total saponins groups.Conclusion:Total saponin of RDN have potential effects on the treatment of gouty arthritis by regulating the content changes of glycine,tryptophan,uric acid,creatinine and lactic acid.
