摘要
目的:观察解毒活血复方(肠复康,CFK)对人结肠癌Lo Vo细胞裸鼠移植瘤血管生成的抑制作用,并探讨其对STAT3、p STAT3及VEGF、MMP-2表达的影响。方法:采用裸鼠结肠癌原位种植转移模型,将荷瘤鼠50只随机分为5组,每组10只,种植后第1周开始,分别给予生理盐水(模型组)、5-FU(5-FU组)、CFK低剂量(CFK低剂量组)、CFK高剂量(CFK高剂量组)、5-FU与CFK联合应用(CFK+5-FU组),1次/d,共用4周。种植后第5周末处死动物,测量原位肿瘤瘤质、抑瘤率,免疫组化检测各组结肠癌组织中肿瘤微血管密度(MVD),Western blot法检测STAT3、p STAT3、VEGF和MMP-2蛋白表达。结果:高剂量组及CFK+5-FU组平均瘤质均显著低于模型组(P<0.05);CFK高剂量组、CFK+5-FU组平均瘤质均低于5-FU组(P<0.05);CFK高、低剂量组、CFK+5-FU组MVD均明显低于模型组和5-FU组,差异均有统计学意义(P<0.01);CFK高剂量组、5-FU组肿瘤组织中STAT3、p STAT3、VEGF及MMP-2蛋白的表达水平均低于模型组,差异均有统计学意义(P<0.01或P<0.05)。结论:解毒活血复方(CFK)可通过干预STAT3磷酸化,下调VEGF、MMP-2蛋白表达,从而抑制人结肠癌Lo Vo细胞裸鼠移植瘤血管生成。
Objective: To observe the inhibition of liedu Huoxue Fufang [Changfukang(肠复康,CFK)]on hu-man colon cancer cells in nude mice LoVo angiogenesis,and explore its effects on the expression of STAT3, pSTAT3, YEGF and MMP-2. Methods: Using 5-fluorouracil (5-FU) as positive control, metastatic model of hu- man colon cancer was established by orthotropic implantation of human tunmr tissue into colon wall of nude mice. A total of 50 mice were randomly divided into five groups, 10 miee in eaeia group, including model con- trol group, 5-FU group, low dose CFK group, high dose CFK group, and combined treatment group (both CFK and 5-FU). Nude miee with cancer cells grown for a week, eac, h mouse was given NS (model control group), 5- FU (5-FU group), low dose CFK (low dose CFK group), high dose CFK (high dose CFK group), both CFK and 5-FU (combined treatment group) respectively everyday for four weeks. The mice were killed in five weeks, and tumor weight, inhibition rates, the expressions of MVD and Application of Western blot assays STAT3, VEGF and MMP-2 expression were detected. Results: The high dose CFK group and combined treatment group showed lower tumor weight than model control group (P〈0.05); The high dose CFK group and combined treatment group showed lower tumor weight than 5-FU group (P〈0.05); The high dose CFK group, low dose CFK group and combined treatment group showed lower MVD than the model control group and 5-FU group (P〈0.01). The high dose CFK group and 5-FU group showed lower STAT3, pSTAT3, VEGF and MMP-2 expression than the con- trol group (P〈0.01 or P〈0.05). Conclusion:Jiedu Huoxue Futang (Changfukang) can inhibit angiogenesis of human colon cancer by preventing STAT3 signaling pathway and reducing the expression of VEGF and MMP-2.
