摘要
目的:建立同时测定活血促愈胶囊中人参皂苷Rg_1、人参皂苷Rb_1、三七皂苷R_1、积雪草苷、羟基积雪草苷含量的方法。方法:采用高效液相色谱法,使用Kromasil 100-5 C_(18)色谱柱,流动相为乙腈-水(梯度洗脱),流速1 m L·min^(–1),检测波长203 nm,柱温30℃。结果:人参皂苷Rg_1、人参皂苷Rb_1、三七皂苷R_1、积雪草苷、羟基积雪草苷阴性均无干扰;人参皂苷Rg_1、人参皂苷Rb_1、三七皂苷R_1、积雪草苷、羟基积雪草苷分别在0.702~7.02μg(r=0.999 6)、0.655 2~6.552μg(r=0.999 8)、0.135 6~1.356μg(r=0.999 9)、0.277 4~2.774μg(r=0.999 9)、0.638 4~6.384μg(r=0.999 9)范围内进样量与峰面积呈良好的线性关系;平均加样回收率(n=6)为97.1%~98.7%,RSD为1.5%~2.7%,3批样品中人参皂苷Rg_1、人参皂苷Rb_1、三七皂苷R_1、积雪草苷和羟基积雪草苷含量范围分别为4.11~4.27、3.82~3.94、0.72~0.84、1.71~1.88和3.10~3.19 mg·g^(-1)。结论:该方法可用于活血促愈胶囊中人参皂苷Rg_1、人参皂苷Rb_1、三七皂苷R_1、积雪草苷、羟基积雪草苷含量的同时测定。
Objective: To establish a method for simultaneous determination of ginsenoside Rgl, ginsenoside Rbl, notoginsenoside R1, asiaticoside and madecassoside in Huoxue Cuyu capsules. Methods: HPLC was performed on the column of Kromasill00-5 Cls with mobile phase of acetonitrile-water (gradient elution )at a flow rate of 1 mL ~ min-1. The detection wavelength was 203 nm and the column temperature was 30℃. Results: No interference was found in negative samples. The linear ranges of ginsenoside Rg1, ginsenoside Rb1, notoginsenoside R1, asiaticoside and madecassoside were 0.702-7.02 μg( r=0.999 6 ), 0.655 2-6.552μg( r=0.999 8 ), 0.135 6-1.356μg ( r=0.999 9 ), 0.277 4-2.774μg ( r=0.999 9 ) and 0.638 4-6.384μg ( r=0.999 9 ), respectively. The average recovery rates were 97.1%-98.7% with RSDs between 1.5% and 2.7% (n=6). The content ranges of ginsenoside Rgl, ginsenoside Rbl, notoginsenoside R1, asiaticoside and madecassoside in samples were 4.11-4.27 -1 -1 - -1 mg·g-1,3.82-3.94mg·g-1 ,0.72-0.84 mg·g-1, 1.71.88mg·g-1 and 3.10-3.19mg·g-1, respectively. Conclusion: The method can be used for the simultaneous determination of ginsenoside Rg1, ginsenoside Rbl, notoginsenoside RI, asiaticoside and madecassoside in Huoxue Cuyu capsules.
