农用抗生素多抗菌素高产菌株的诱变及发酵优化 被引量:2

摘要 利用N+注入技术对出发菌株金色产色链霉菌(Streptomyces aureochromogenes)NJYHWG66302进行诱变选育,考察不同注入能量和剂量对菌株存活率和正负突变率的影响。得出注入诱变的最佳条件为:16keV,24×1014 ions/cm2和18keV,20×1014ions/cm2。在最佳注入条件下,对出发菌株经过多轮诱变选育,筛选得到1株多抗菌素高产菌株并命名为YX1,较出发菌株多抗菌素效价提高了约36%,平均效价可达2 828mg/L,并能稳定遗传7代以上。以YX1为研究对象,对其发酵培养基及培养条件进行探究,并考察碳源、无机盐、种龄、pH及装液量对多抗菌素发酵的影响。最终确定最佳发酵配方为:酵母膏,18.72g/L;蔗糖,17.42g/L;KH2PO4,0.69g/L;FeSO4,0.2g/L;CaCO3,0.3g/L,最适装液量为50mL,最适接种量为5%,最适初始pH值为6.0,发酵平均效价可达3394mg/L,与为优化前相比提高了20.7%。 The N+injection mutagenesis has been used to mutagenize the strain Streptomyces aureochromogenes NJYHWG66302 and the influence of different implantation energies and doses on the survival rate of strains mutation rate has been investigated.The optimal conditions are as follows:16 keV,24×1014 ions·cm-2 and 18 keV,20×1014 ions·cm-2.Under these conditions,a strain was screened and named YX1.Compared with the original strain,the antibiotic titer was increased by about 36%,the average titer was 2828 mg/L,and the genetic stability could be inherited over 7 generations.Taking YX1 as the research object,the fermentation medium and culture conditions were investigated,and the effects of carbon source,inorganic salt,optimum age,initial pH and liquid content on the fermentation of polyoxins were investigated.The final fermentation formula was determined as following:Yeast extract is 18.72 g/L,sucrose is 17.42 g/L,KH2 PO4 is 0.69 g/L,FeSO4 is 0.2 g/L,CaCO3 is 0.3 g/L.The optimum dosage is 50 mL,the optimum inoculation amount is 5%,the optimum initial pH is 6.0,the average titer of fermentation is 3394 mg/L,which is 20.7% higher than that before optimization.
