Effects of hepatocyte growth factor-regulated tyrosine kinase substrate on the expression and secretion of insulin

摘要 目的探讨肝细胞生长因子调节的酪氨酸激酶底物(HGS)对胰岛素表达和分泌的影响。方法检测HGS,胰岛素和胰升血糖素在小鼠胰岛中的分布。检测不同浓度葡萄糖(5.5、11.1及16.7mmol/L)培养24h后NIT-1细胞HGS表达水平和胰岛素的分泌。观察siRNA沉默HGS表达后48h NIT-1细胞胰岛素的表达和分泌的变化。结果 HGS免疫阳性细胞特异性分布于小鼠胰腺的胰岛,并与胰岛素共定位于胰岛β细胞中,未见HGS和胰高血糖素阳性荧光双标细胞。不同浓度葡萄糖培养NIT-1细胞24h,5.5mmol/L组为(25.07±0.93)μIU/mg,11.1mmol/L组为(32.28±0.93)μIU/mg,16.7mmol/L组为(41.77±1.39)μIU/mg,NIT-1细胞分泌胰岛素水平逐渐增加(P<0.01)。各组HGS蛋白水平也随葡萄糖浓度增加而逐渐增加(P<0.05)。siRNA沉默HGS表达后NIT-1细胞内胰岛素的表达水平无明显变化,但胰岛素分泌水平减少(P<0.05)。结论 HGS选择性表达于胰岛β细胞中,并调节胰岛素的分泌。 Objective To evaluate the effects of hepatocyte growth factor-regulated tyrosine kinase substrate(HGS)on the expression and secretion of insulin. Methods The distributions of HGS,insulin and glucagon in pancreatic islets of mice were examined.The levels of HGS protein and insulin secretion in NIT-1 cells were detected after culturing in different concentrations of glucose(5.5 mmol/L,11.1 mmol/L and 16.7 mmol/L)for 24 hours.Furthermore,we observed insulin expression and secretion in NIT-1 cells after silencing HGS for 48 hours. Results HGS-positive cells were specifically distributed in mice islets.Colocalization of HGS with insulin but not glucagon was observed in islet beta cells.The levels of insulin secretion in groups of 5.5 mmol/L glucose[(25.07±0.93)μIU/mg],11.1 mmol/L glucose[(32.28±0.93)μIU/mg]and 16.7 mmol/L glucose[(41.77±1.39)μIU/mg]were gradually increased in NIT-1 cells after culturing for 24 hours(P〈0.01).The levels of HGS protein in these 3 groups also gradually increased with the rise of glucose concentration(P〈0.05).No significant difference of insulin expression was found in NIT-1 cells between groups of HGS siRNA and negative control[(102.6±3.32)vs(98.38±3.87)μIU/mg,P〈0.05].However,insulin secretion was significantly decreased in HGS siRNA group[(32.58±2.74)vs(23.18±1.79)μIU/mg,P〈0.05]. Conclusion HGS is selectively expressed inβcells of pancreatic islets,and regulates the secretion of insulin.
