摘要 目的 克隆并表达幽门螺杆菌粘附素 Alp A基因 .方法 提取幽门螺杆菌染色体基因 ,用 PCR方法扩增 alp A基因 ,将其克隆至表达载体 p ET- 2 2 b (+) ,转化大肠杆菌BL 2 1 (DE3) ,IPTG诱导表达 .结果 分离得到了 1 .5 kb的alp A基因片段 ,并在大肠杆菌中实现了该基因的高效表达 ,在 37℃诱导表达 3h后 ,表达产物占细菌总蛋白的 31 .9% .表达以可溶性蛋白和包涵体两种形式存在 ,其中主要是包涵体的形式 ,目的蛋白占不溶性蛋白的 64.8% .结论 幽门螺杆菌 alp A基因的克隆与表达为 AIM To isolate and expresse alpA gene from Helicobacter pylori . METHODS The alpA gene was amplified from H.pylori chromosomal by PCR and inserted into the prokaryotie expression vector pET 22b(+).The recombinant plasmid was transformed into the BL21 (DE3) E.coli strain. RESULTS 1.5 kb alpA gene was successfully isolated. Recombinant E.coli strains expressed AlpA were obtained, the expression protein amounted to 31.9% of the total bacterial protein after induced with IPTG for 3 h at 37℃. CONCLUSION Cloning and high expression of alpA gene lay the foundation for the development of the H.pylori vaccine.
