四乙酰基二苄基六氮杂异伍兹烷致大鼠急性经口毒性研究

摘要 目的研究TADB对大鼠的急性经口毒性,检测化学品对实验动物的急性毒性作用和强度,为亚急(慢)性毒性试验提供剂量选择的依据。方法将30只成年SPF级SD大鼠按体重随机分为2组:实验组20只,给予250mg/ml的TADB混悬液;对照组10只,给予一定量的植物油。采用经口灌胃方式进行染毒,观察染毒后14 d内动物的反应、体重变化和死亡情况。实验结束后对其心、肝、脾、肺、肾等脏器进行病理检查。结果实验组和对照组在观察期内均未出现中毒症状和死亡,且染毒组实验动物体重增长速度与对照组相比差异无统计学意义(P>0.05)。病理检查结果显示实验组大鼠出现中度肝细胞肿胀,伴有肝窦及中央静脉扩张充血,其他脏器均无明显病理改变。结论 TADB的雌、雄性大鼠LD50均>5 000 mg/kg,属于微毒物质。 Objective To study the acute oral toxicity of Etraacetyldibenzulhexaazaisowurtzitane( TADB) in rats,to detect the acute toxic effects and the density of the chemical in rats,in order to provide the reference dose for sub-acute( sub-chornic) toxicity experiment.Methods 30 adult SD rats were randomly divided into two groups by body weights,with 20 in dose group and 10 in control group. The rats in dose group were fed with 250 mg/ml TADB suspension,while vegetable oil in control group. The general performance,poisoning symptoms and death were observed during the 14 days. At the end of the 14 th days,routine pathological examinations for heart,liver,spleen,lung and kidney were carried out. Results None poisoning symptoms and death were observed during the experiment,while there was no differences between the increasing rate of body weight in dose group and the control group in both male and female rats. The moderate liver cell swelling was observed in dose group. The histological and pathological examination showed that the rats in the experiment group had moderate hepatocyte swelling,accompanied by expansion and hyperemia of hepatic sinus and central venous. However,no obvious abnormality was found in the pathological examination of other viscera.Conclusion The LD50 of TADB in male and female rats were both more than 5 000 mg/( kg·bw) which belongs to mild toxicity.
