摘要
目的:探究白细胞介素22(interleukin-22,IL-22)在细胞增殖以及细胞因子表达方面对人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)的影响。方法:用RT-PCR技术检测IL-22R1在HUVECs中的表达情况。将以脂多糖(Lipopolysaccharide,LPS)处理后的HUVECs作为血管炎细胞模型,CCK8实验用于检测IL-22作用前后HUVECs的增殖能力变化情况,q RT-PCR技术以及Western blot技术则分别用于检测IL-22作用前后IL-6,IL-8,CC类趋化因子配体2(chemokine motif lig and 2,CCL2),细胞间粘附分子1(intercellular cell adhesion molecule-1,ICAM1),血管细胞粘附分子-1(vascular cell adhesion molecule 1,VCAM1),趋化因子受体4(chemokine receptor 4,CXCR4)和CCL20等细胞因子在HUVECs中mRNA水平以及蛋白质水平的表达变化情况。结果:实验结果显示IL-22R1 mRNA在HUVECs中广泛表达。IL-22作用于HUVECs后,细胞的增殖率得到明显提高。此外,LPS(脂多糖)可以显著抑制HUVECs的增殖率,而被IL-22预处理过的HUVECs其增殖率不受LPS的抑制。在IL-22作用于HUVECs后,趋化因子CCL2和CCL20的表达在mRNA水平及蛋白质水平均可见明显增加,而炎症因子IL-6及IL-8的表达则未见明显变化。结论:IL-22的受体IL-22R1在HUVECS中广泛表达。IL-22不仅可以提高HUVECs的增殖能力,还可以抑制由LPS诱导的增殖抑制。此外,IL-22还可以提高趋化因子CCL2与CCL20的表达,却对炎症因子IL-6及IL-8的表达无明显影响。由此可以推测IL-22在血管炎的发展过程中具有抗炎的作用。
Objective:To investigate the effects of interleukin-22(IL-22)on proliferation function and cytokine production of HUVECS.Methods:The expression of IL-22 R1 mRNA in HUVECS was detected by semiquantitative reverse transcription-polymerase chain reaction(RT-PCR).The proliferation ability of HUVECS was evaluated using a cell counting kit assay.Real-time quantitative PCR technique and Western blot were used to detect the expression of cytokine.Results :IL-22 R1 mRNA expression was noted in HUVECS.After treatment with IL-22,the HUVECS showed increased proliferation activity,and IL-22 could effectively suppress LPS-induced proliferation inhibition.IL-22 also significantly enhanced the production of CCL2 and CCL20 by HUVECS,but exerted no effect on expression of IL-6 and IL-8.Conclusion:IL-22 could not only significantly enhance the proliferation ability of HUVECS,but also enhance the production of CCL2 and CCL20,which known as chemotactic cytokines by HUVECs.IL-22 exerts no effect on expression of IL-6 and IL-8,which known as inflammatory factors.These results suggest that IL-22 may play a protective role in vasculitis.
