柿叶提取物减轻大鼠心梗后心肌细胞凋亡和心室重塑相关机制研究 被引量:7

Alleviation of persimmon leaf extract on post-infarction cardiomyocyte apoptosis and myocardial remodeling in rats

摘要 目的:探讨大鼠心梗模型下柿叶提取物对心室重塑的影响及相关机制。方法:用24只Sprague Dawley大鼠结扎左冠状动脉前降支制作心肌梗死大鼠模型,随机分为心肌梗死组(n=8)、柿叶提取物组(n=8)、假手术组(n=8)。6周后,用TUNEL法测定心肌细胞凋亡,免疫印迹法检测心肌Fas、Caspase-3蛋白含量,并测定大鼠血流动力学参数和左心室质量指数。结果:柿叶提取物组、心肌梗死组左心室质量指数分别为2.417±0.053、2.514±0.101(F=136.762,P=0.011),明显低于心肌梗死组;柿叶提取物组、心肌梗死组Fas分别是0.342±0.058、0.585±0.059(F=163.991,P=0.000);柿叶提取物组、心肌梗死组Caspase-3蛋白水平分别是0.124±0.019、0.478±0.079(F=183.553,P=0.000),明显低于心肌梗死组。柿叶提取物组、心肌梗死组心肌细胞凋亡指数分别是1.198±0.171、0.507±0.132(F=93.023,P=0.000),明显低于心肌梗死组。柿叶提取物组左心室舒张末期压(left ventricular end diastolic pressure,LVEDP)明显降低(F=347.661,P=0.000),左心室收缩压(left ventricular systolic pressure,LVSP)明显增加(F=444.949,P=0.000)。结论:心肌细胞凋亡参与心肌梗死后心室重塑过程,柿叶提取物可能通过减少凋亡相关蛋白表达抑制心肌细胞凋亡从而预防心室重塑,改善心肌梗死后心脏收缩功能。 Objective:To investigate the effect and mechanism of persimmon leaf extract on apoptosis-related protein dependent signal pathway mediated myocardial remodeling after myocardial infarction in rats.Methods:Twenty-four Sprague Dawley rats were established as myocardial infarction model by ligating the left anterior descending coronary artery and were randomized into three groups:myocardial infarction group(n=8),persimmon leaf extract administration group(n=8),sham-operated group(n=8).After six weeks,cardiomyocyte apoptosis was measured by TUNEL,Fas and Caspase-3 protein expression was detected by Western blot and hemodynamics and left ventricular mass index(LVMI)of rats were measured.Results:The LVMI in the persimmon leaf extract group(2.417±0.053)was lower than that in the myocardial infarction group(2.514±0.101)(F=136.762,P=0.011).The Fas in the persimmon leaf extract group and the myocardial infarction group were 0.342±0.058,0.585±0.059,respectively(F=163.991,P=0.000).The level of Caspase-3 protein in the persimmon leaf extract group(0.124±0.019)was significantly lower than that in the myocardial infarction group(0.478 ±0.079)(F =183.553,P =0.000).The index of myocardial apoptosis in the persimmon leaf extract group(1.198±0.171)was significantly lower than that in the myocardial infarction group(0.507±0.132)(F=93.023,P=0.000).In the persimmon leaf extract group,the LVEDP was decreased significantly(F=347.661,P=0.000)and the LVSP was increased significantly(F=444.949,P=0.000).Conclusion:Cardiomyocyte apoptosis is involved in myocardial remodeling after myocardial infarction.Persimmon leaf extract can prevent myocardial remo-deling and improve post-infarction systolic function by reducing the expre-ssion of apoptosis-related protein and inhibiting the cardiomyocyte apoptosis.
