摘要
为了探索FOXL2和FIGLA基因与绵羊产羔数之间的关系,利用荧光定量PCR检测了二者在多羔小尾寒羊和单羔小尾寒羊8个重要繁殖相关组织中的表达水平。结果表明:FOXL2和FIGLA基因均在卵巢组织中高表达,其次在下丘脑、垂体、输卵管、子宫体以及子宫角中呈中等丰度表达,在大脑和小脑组织中表达量相对较低。其中,FOXL2基因在多羔小尾寒羊小脑和子宫体中的表达量显著高于单羔组(P<0.05),但在多羔小尾寒羊垂体和卵巢中的表达量显著低于单羔组(P<0.05),在多羔小尾寒羊下丘脑中的表达量极显著低于单羔组(P<0.01);FIGLA基因在多羔小尾寒羊大脑中的表达量显著高于单羔组(P<0.05),在其它各组织间表达差异均不显著(P>0.05)。研究初步表明,FOXL2基因表达可能对绵羊产羔数有一定影响,FIGLA基因表达对绵羊产羔数没有影响。
In order to explore the association of FOXL2 and FIGLA with litter size in sheep,real-time PCR was performed to detect FOXL2 and FIGLA mRNA expression in eight important reproduction related tissues of both multiparous and uniparous Small Tail Han ewes.The results showed that the expression of FOXL2 and FIGLA was at high level in ovary of both multiparous and uniparous Small Tail Han Sheep,followed by hypothalamus,pituitary,oviduct,uterus body and uterin horn.The expression was relatively low in brain and cerebellum.The expression of FOXL2 in cerebellum and uterus body in multiparous sheep was significantly higher than that in uniparous sheep(P<0.05).The expression of FOXL2 in pituitary and ovary in multiparous sheep was significantly lower than that in uniparous sheep(P<0.05).The expression of FOXL2 in hypothalamus in multiparous sheep was significantly lower than that in uniparous sheep(P<0.01).The expression of FIGLAin brain of multiparous sheep was significantly higher than that of uniparous sheep(P<0.05),and there were no significant differences in other tissues between multiparous and uniparous sheep(P>0.05).This study preliminarily indicated that the FOXL2 expression may influence sheep litter size to some extent,while the FIGLAexpression may have no effect on sheep litter size.
