摘要
目的 :研究千金子生品和霜品对大鼠小肠黏膜中的乳酸脱氢酶(LDH)、苹果酸脱氢酶(MDH)和乳糖酶3种消化酶作用的差异。方法 :将56只Wistar大鼠随机分成空白对照组、生品高剂量组(23.3529 g·kg^-1)、生品中剂量组(11.6764 g·kg^-1)、生品低剂量组(5.8382 g·kg^-1)、霜品高剂量组(23.3529 g·kg^-1)、霜品中剂量组(11.6764 g·kg^-1)、霜品低剂量组(5.8382 g·kg^-1),每组8只。对照组给予生理盐水,其余6个给药组分别灌胃千金子制霜前后提取物高、中、低剂量的混悬液,每天给药1次,连续给药4 d,取材并处理后采用比色法检测大鼠小肠黏膜LDH、MDH和乳糖酶活性。结果 :千金子生品和霜品均会降低大鼠小肠黏膜LDH、MDH和乳糖酶的活性(P〈0.05),且酶的活性随着给药剂量的增大而减小,相同剂量下,千金子霜品实验组大鼠的3种肠道酶活性大于千金子生品实验组大鼠的3种肠道酶活性(P〈0.05)。结论 :千金子生品和霜品可以降低小肠3种消化酶的活性,且与给药剂量有一定的关系,同种剂量下,千金子制霜后对肠道酶活性的降低程度小于生品,表明制霜后毒性降低,从肠道酶的角度可以证明千金子制霜减毒的科学性。
Objective :To study the diffefences between the effects of Semen Euphorbiae and Semen Euphorbiae Pulveratum on the LDH,MDH and lactase of intestinal mucosa of rats. Methods :Fiftysix Wistar rats were randomly divided into blank control group,high dose Semen Euphorbiae group(23.3529 g·kg^-1),middle dose Semen Euphorbiae group(11.6764 g·kg^-1),low dose Semen Euphorbiae group(5.8382 g·kg^-1),high dose Semen Euphorbiae Pulveratum group(23.3529 g·kg^-1),middle dose Semen Euphorbiae Pulveratum group(11.6764 g·kg^-1),low-dose Semen Euphorbiae Pulveratum group(5.8382 g·kg^-1)group,each included 8 rats. The rats in the control group were treated with normal saline,and the other six groups were given high,medium and low dose of the extracts from Semen Euphorbiae and Semen Euphorbiae Pulveratum. The rats were given once a day for 4 days,LDH,MDH and lactase activities of small intestine mucosa were measured by colorimetry. Results :The activities of LDH,MDH and lactase of intestine mucosa of rats were decreased(P〈0.05),and the activity of enzyme decreased with the dose increasing. At the same dose,The activities of three intestinal enzymes in the experimental group were higher than those in the experimental group(P〈0.05). Conclusion :Semen Euphorbiae and Semen Euphorbiae Pulveratum can reduce the activity of three kinds of digestive enzymes in intestine and have a certain relationship with the dose. Under the same dose,the decrease of intestinal enzyme activity of Semen Euphorbiae Pulveratum is less than Semen Euphorbiae,indicating that after the reduction of toxicity after the preparation of Semen Euphorbiae Pulveratum,from the perspective of intestinal enzymes can prove scientific toxicity reduction of Semen Euphorbiae Pulveratum.
