针刺与穴位埋线对穴区胶原纤维的影响 被引量:4

The Effects of Acupuncture and Catgut Implantation on Acupoint Collagen Fiber

摘要 目的:对比针刺与穴位埋线在不同干预时间内对大鼠肝俞穴区胶原纤维变化的影响。方法:选取56只清洁级SD大鼠,随机分为空白组、针刺组和穴位埋线组,按照干预时间将针刺组和穴位埋线分为4天、7天和14天3个时间段亚组,每组8只。针刺组每日针刺右侧肝俞一次,深度至皮下4mm。穴位埋线组采用简易穴位埋线法将4mm长的可吸收缝合线埋入右侧肝俞穴。空白组予针刺组和穴位埋线组相同的麻醉、抓取和固定,不做其他处理。在干预4天、7天和14天后处死大鼠,取出右侧肝俞穴区肌肉组织进行HE染色、MASSON三色改良染色。镜下观察穴区深部肌纤维、胶原纤维形态与量的变化。结果:干预4天后,针刺组穴区胶原纤维较空白组与穴位埋线组均有一定程度的增加,有统计学意义(P<0.05)。干预7天、14天后,针刺组和穴位埋线组与空白组胶原纤维比较均具有显著性差异(P<0.01)。针刺4天、7天、14天后,针刺组穴区胶原纤维随着干预时间的延长而增多,其中4天组和14天组比较有统计学意义(P<0.05)。穴位埋线4天、7天、14天后,穴区胶原纤维也随着干预时间的延长而增多,7天、14天组与4天组比较具有显著性差异(P<0.01)。结论:针刺和穴位埋线在14天内随着干预时间的延长均可使穴区深部组织的胶原纤维增多。针刺组增生的胶原纤维主要在肌纤维变形、扭曲与断裂处,穴位埋线增生的胶原纤维主要沿着肌纤维长轴方向走行,分布较均匀。 Objective To compare the effects of acupuncture and catgut implantation on changes of the collagen fibers in Ganshu Points of rats at different intervention time. Method: 56 clean grade SD rats were randomly divided into a blank group, an acupuncture group and a catgut implantation group. The acupuncture group and the catgut implantation group were subdivided into 4-day, 7-day and 14-day intervention subgroups respectively with 8 rats in each group. Acupuncture was operated subcutaneously in the right Ganshu Points by the depth of 4mm once a day in the acupuncture group. 4mm long absorbable sutures were embedded in the same point in the catgut implantation group. The same anesthesia, grabbing and fixing treatments were performed to the blank group with no extra intervention. Rats were sacrificed on the 4th, 7th and 17th day of intervention and the muscle tissues of the right Ganshu Points were taken for HE staining and the improved MASSON staining. The changes of morphology and deep muscle and collagen fibers were observed under microscope. Results: A certain degree of increasing of connective tissue was observed in the acupuncture group after 4 day' s intervention compared to the blank group and the catgut implantation group (P〈0.05). After 7 day' s and 14 day' s intervention, there were significant difference of Collagen fibers in the acupuncture group and the catgut implantation group compared with the blank group (P〈0.01). After 4 day' s, 7 day' s and 14 day' s intervention, collagen fibers were increased in the acu- puncture group with the prolongation of the intervention time. There were significant differences between 4-day and 14-day subgroups (P〈0.05). After 4 day' s, 7 day' s and 14 day' s intervention, collagen fibers were also increased in the catgut implantation group with the prolongation of intervention time. A significant difference was observed in the 7-day and the 14-day subgroups compared with the 4-day subgroup (P〈0.01). Conclusion: Collagen fibers were significantly increased along with the extension of the intervention time within 14 days in both the acupuncture and the catgut implantation group. The distribution of increased collagen fibers were mainly in the places of muscle fiber deformation, distortion and fracture in acupuncture group whereas the distribution in the catgut implantation group was even along the direction of the long axis of muscle fibers.
