摘要
目的 观察复方黄连消疮灵抗炎作用,探讨其对复发性口腔溃疡(recurrent oral ulceration, ROU)模型大鼠炎症的影响。方法 将60只小鼠按随机数字表法分为空白组,模型组,复方黄连消疮灵高、中、低剂量组(2、1、0.5 g/kg生药量),布洛芬组(0.15 g/kg),利用小鼠耳二甲苯致炎试验,观察其抗炎作用;采用免疫法建立大鼠ROU模型,将60只成模SD大鼠按随机数字表法分为正常对照组,模型对照组,复方黄连消疮灵高、中、低剂量组(1、0.5 、0.25 g/kg生药量),左旋咪唑组(20 mg/kg),各组均灌胃给予相应药物,模型对照组、正常对照组灌胃给予等容量纯水,共30 d,观察溃疡数目、持续时间和间隔时间,及对血清IL-4、IL-8、VEGF表达水平的影响。结果 与二甲苯致炎小鼠模型组比,复方黄连消疮灵高、中剂量小鼠耳肿胀度[(7.46±1.41)mg、(9.85±2.71)mg比(16.25±3.34)mg]减少(P<0.05);与ROU大鼠模型对照组比较,复方黄连消疮灵高、中剂量大鼠溃疡数目[(4.70±1.27)、(6.90±3.70)比(9.90±5.13)]减少(P<0.05),溃疡持续时间[(2.20±1.22)d、(3.00±1.05)d比(5.00±1.15)d]缩短(P<0.05),溃疡间隔时间[(8.00±1.63)d、(5.00±1.82)d比(2.40±1.26)d]延长(P<0.05)。复方黄连消疮灵高剂量组血清IL-4[(68.92±10.58)pg/ml比(55.18±6.99)pg/ml]含量升高(P<0.05),血清IL-8[(399.86±62.14)ng/ml比(578.70±72.23)ng/ml]含量降低(P<0.05);高、中剂量组VEGF[(41.41±3.80)pg/ml、(37.92±3.48)pg/ml比(32.01±4.32)pg/ml]含量升高(P<0.05)。结论 复方黄连消疮灵有抗炎作用,可通过上调血清IL-4和VEGF表达、下调血清IL-8表达,治疗ROU。
Objective To study the mechanism of Compound berberine Xiaochuang powder on recurrent oral ulcers,and the anti-inflammatory effect. Methods The mice were divided into blank group, model group, and compound yellow hyalensis with high, medium, low dose group(2, 1, 0.5 g/kg equivalent medicine) and ibuprofen group(0.15 g/kg). Using mouse ear inflammation test by Xylene, To establish recurrent oral ulcer model by immunoassay in Rats, the mold SD rats according to random number table method is divided into blank group, model group, compound coptis sores spirit high, medium and low dose group (1, 0.5, 0.25 g/kg equivalent medicine), levamisole group (20 mg/kg), each group all gastric gavage corresponding drugs, model group, the blank group gastric gavage capacity such as pure water, a total of 30 days, observe the number of ulcers, duration and interval time, and the effection on serum IL-4, IL-8, VEGF level. Results The compound berberine Xiaochuang powder high dose may significantly reduce the swelling degree of ears (7.46 ± 1.41 mg, 9.85 ± 2.71 mg vs. 16.25 ± 3.34 mg, P〈0.05), reducing the number of ulcers (4.70 ± 1.27, 6.90 ± 3.70 vs. 9.90 ± 5.13, P〈0.05), reduce ulcer duration (2.20 ± 1.22 d, 3.00 ± 1.05 d vs. 5.00 ± 1.15 d, P〈0.05), prolonged intervals ulcers (8.00 ± 1.63 d, 5.00 ± 1.82 d vs. 2.40 ± 1.26 d, P〈0.05). Compound berberine Xiaochuang powder high dose can be significantly increased the level of serum IL-4 (68.92 ± 10.58 pg/ml vs. 55.18 ± 6.99 pg/ml,P〈0.05) and lowered serum IL-8 (399.86 ± 62.14 ng/ml vs. 578.70 ± 72.23 ng/ml,P〈0.05); The compound berberine Xiaochuang powder high dose could significantly increase the expression of VEGF (41.41 ± 3.80 pg/ml、37.92 ± 3.48 pg/ml vs. 32.01 ± 4.32 pg/ml,P〈0.05). Conclusions The compound berberine Xiaochuang powder can increased the level of serum IL-4 and VEGF, reduced the level of IL-8, so as to achieve the treatment of recurrent oral ulcers.
