摘要
【目的】探索铁冬青组织培养的可行性,便于铁冬青的组培育苗生产。【方法】以3年生铁冬青茎段为外植体,研究MS基本培养基下,不同质量浓度的细胞分裂素和生长素的配比对铁冬青芽诱导的影响,及在含不同质量浓度的MS培养基、细胞分裂素和生长素下对铁冬青芽的继代增殖效果。【结果】铁冬青芽诱导的MS培养基,高浓度的6-BA能有效缩短萌芽的时间且有一定量的增殖,IAA对萌芽的生长有一定的影响,但没有6-BA效果明显;铁冬青芽的增殖培养基中,1/2MS培养基含高浓度细胞分裂素和生长素的增殖系数有明显差异。在MS培养基中,激素影响增殖效果并不显著。【结论】铁冬青芽诱导的培养基可采用MS+6-BA 1mg/L+IAA 0.2mg/L;在考虑生产成本的前提下,增殖培养基采用1/2MS+6-BA 1.5mg/L+IAA 0.5mg/L,增殖系数为3.3。
【Objective】It aims to explore the feasibility of tissue culture of Ilex Rotunda and to facilitate the production of tissue culture seedling.【Method】On the basis of MS medium,3-year-old pig iron holly stem is considered as explants to study the influence of the ratio of cytokinin & auxin with different concentrations and Ilex rotunda bud subculture propagating effect induced by different concentrations of MS medium & cytokinin & auxin.【Result】Induced iron Holly buds on MS medium and high concentration of 6-BA can effectively shorten the germination time with a certain amount of propagating.IAA has certain influence on the germination,but the effect is not as obvious as 6-BA; in the iron Holly buds propagating medium,1/2 MS culture medium's propagating coefficients of high concentrations of cytokinin and the auxin have obvious differences. In MS medium,the effect of hormone on propagating was not significant.【Conclusion】Rotundic bud induction medium can use MS+6-BA 1 mg/L+IAA 0. 2 mg/L; considering the premise of production cost,propagating medium can adopt 1/2 MS + 6-BA 1. 5 mg/L + IAA 0. 5 mg/L,with the propagating coefficient 3. 3.
