摘要
目的观察分析尼古丁对大鼠急性心肌梗死后心肌瘢痕形成的影响,并初步探讨其发生的可能机制。方法参考Lobo等急性心梗模型方法建立大鼠急性心肌梗死模型,经腹腔注射尼古丁干预心肌瘢痕的形成,苏木精-伊红(H—E)染色观察心肌组织病理变化,ELISA检测心肌肌钙蛋白(cTN—T)含量变化,氯胺T法检测梗死区胶原含量。结果实验组与对照组比较,大鼠心肌梗死后注射尼古丁后,血清cTN—T含量明显升高,心肌羟脯氨酸含量明显降低,差异有统计学意义。结论尼古丁对心肌梗死后心肌瘢痕形成有一定抑制作用,可能与尼古丁影响成纤维细胞增殖分化有关。
Objective To observe the effects of nicotine on the formation of myocardial scar in rats with acute myo- cardial infarction and explore its mechanism. Methods A rat model of myocardial infarction was established through the Lobo method. Then, nicotine was intraperitoneally injected into to interfere with the formation of myocardial scar. The pathological changes in myocardial tissues were observed using hematoxylin eosin (HE) staining. The content of cardiac troponin ( cTN - T) was measured by ELISA. The level of myoeardium collagen in the infarction area was detected by the chloramine T method. Results Compared with the control, remarkably increased levels of serum cTN - T and markedly decreased amounts of hydroxyproline were found in myocardial infarction rats after injection of nicotine. Conclusion Nicotine may inhibit the formation of myocardial scar after myocardial infarction, which may be related to its inhibitory role of the proliferation and differentiation of fibroblasts.
