摘要
5-脂氧合酶(5-lipoxygenase,5-LOX)是催化花生四烯酸(arachidonic acid,AA)生成5-羟基-6,8,11,14-二十碳四烯酸(5-hydroperoxy-6,8,11,14-eicosatetraenoic acid,5-HPETE)和各种白三烯(leukotrienes,LTs)类物质的关键酶。近年来研究显示,5-LOX参与阿尔茨海默病(Alzheimer’s disease,AD)的病理生理过程。抑制5-LOX可以降低β淀粉样蛋白(β-amyloid peptide,Aβ)的沉积、tau蛋白磷酸化、减轻神经炎症反应和调节突触功能,改善AD的认知功能障碍。该文就5-LOX与AD相关性研究进展进行综述。
5-1ipoxygenase plays a key role in catalyzing the conversion of arachidonic acid (AA) to 5-hydroperoxy-6,8,11,14-eicosatetraenoic acid (5-HPETE), which can then be metabolized into different leukotrienes (LTs). The recently studies have shown that 5-1ipoxygenase is involved in the pathophysiology of Alzheimer's disease. Inhibition of 5-1ipoxygenase decreases amyloid ]3 peptide (AI3) and tau hyperphosphorylation,mitigates neuroinflammation,regulates synaptic function and ameliorates cognitive disorders in Alzheimer's disease. In this review,we will summarize studies of 5-1ipoxygenase in Alzheimer's disease pathogenesis.
