聚酰胺树脂纯化富集沉香叶中主要黄酮苷元的工艺 被引量:4

摘要 目的利用单因素试验法优选沉香叶中4种主要的黄酮苷元成分的提取工艺。方法采用单因素实验设计,以沉香叶中主要的黄酮苷元成分芫花素、芹菜素、羟基芫花素和木犀草素为对照成分,利用高效液相色谱法测定4种主要黄酮苷元的含量。结果 60~100目聚酰胺树脂为纯化沉香叶4种主要黄酮苷元的最优聚酰胺树脂类型,最佳工艺为,上样流速2BV·h^(-1)(柱体积Bed Column,简称BV),上样液浓度0.09mg·m L^(-1),最大上样量为100BV(178.4mg),最佳洗脱剂为30%乙醇除杂,50%乙醇洗脱,洗脱体积为24BV,50%乙醇洗脱段4种黄酮苷元平均纯度为51.37%,达到中药有效部位要求,4种主要黄酮苷元成分总提取率高达84%。结论为后期合理提取纯化富集沉香叶中黄酮苷元类成分的药用资源提供了工艺参考。 OBJECTIVE To optimize extraction and purification technology of 4 main flavonoids from Aquilaria sinensis Leaves by polyamide resin. METHODS To develop a HPLC method for determination of 4 major flavonoid components,including genkwanin,apigenin,luteolin,hydroxygenkwanin. Absorption and elution behaviors of polyamide resin for the 4 flavonoids was investigated by absorption-elution test and signal factor test. RESULTS The optimized technology parameters were as follow: sample flow rate was 2 BV ·h^(-1); concentration of sample solution was0. 09 mg·m L^(-1); maximum sample volumn was 100 BV( 178. 4 mg); impurities elution was 30% ethanol,then eluted with 24 BV of 50% ethanol. The average purity of 4 flavonoids was 51. 37%. The extraction rate of 4 flavonoids was84%. CONCLUSION This study could provide a good extraction and purification medthod for Flavonoids in Aquilaria sinensis Leaves.
