高效液相色谱法同时测定炒防风中升麻素苷和5-O-甲基维斯阿米醇苷含量 被引量:11

摘要 目的建立同时测定炒防风中升麻素苷和5-O-甲基维斯阿米醇苷含量的高效液相色谱法。方法色谱柱为Rigol-C_(18)柱(250 mm×4.6 mm,5μm),流动相为甲醇-水(40∶60),流速为1.0 mL/min,进样量为5μL,检测波长为254 nm,柱温为35℃。结果可在20 min内完成对升麻素苷和5-O-甲基维斯阿米醇苷的色谱分析,升麻素苷和5-O-甲基维斯阿米醇苷的质量浓度线性范围分别为0.0116~0.2920 g/L(r=0.9994)和0.0114~0.2850 g/L(r=0.9994),平均回收率分别为95.82%和107.70%,RSD分别为2.92%和2.45(n=6)。结论该法方便、快捷、准确、简便、重复性好,可用于炒防风的质量控制。 Objective To establish an HPLC method for simultaneous determination of prim-O-glucosylcimifugin and 5-O-methylvisammioside in fried Radix Saposhnikovia divaricata. Methods The Rigol-C18 column( 250 mm × 4. 6 mm,5 μm) was adopted,the mobile phase was methanol-water( 40 ∶ 60),the flow rate was 1. 0 mL/min,the sample size was 5 μL,the detection wavelength was set at 254 nm,and the column temperature was 35 ℃. Results The chromatographic analysis of prim-O-glucosylcimifugin and 5-O-methylvisammioside were completed within 20 min. Prim-O-glucosylcimifugin and 5-O-methylvisammioside showed good linear relationship within the ranges of 0. 0116-0. 2920 g/L( r = 0. 9994) and 0. 0114-0. 2850 g/L( r = 0. 9994), the average recoveries were 95. 82% and 107. 70% and RSDs were 2. 92% and 2. 45%( n = 6). Conclusion The method is convenient,quick,accurate,simple and reproducible,which can be used for quality control of fried Radix Saposhnikovia divaricata.
