摘要
目的:研究金匮肾气丸对增龄过程中小鼠脾组织代谢物的变化,分析与12月龄雄性小鼠密切相关的生物标记物及其代谢通路,进一步阐释金匮肾气丸调节作用的生物学基础。方法:取3月龄雄性小鼠10只为空白组,12月龄雄性小鼠20只,随机分为模型组和金匮组,金匮组给药金匮肾气丸汤剂,空白组和模型组给予生理盐水,灌胃给药30日后处死小鼠,取脾组织进行实验研究。结果:得到与生长发育相关的内源性代谢差异物26个,如Xanthine、Pyruvic acid、Threonic acid、Glycocholic acid等。其中11个标记物表现为上调,15个标记物表现为下调。主要涉及三条代谢通路。发现金匮肾气丸组对其中9个生物标记物有明显的调节作用。结论:通过代谢组学研究,发现金匮肾气丸通过调节牛磺去氧胆酸、甘氨胆酸、黄嘌呤等标记物,对小鼠的增龄起到了干预作用。
Objective: To study the effect of Jingui Shenqi pill on the metabolites changes of spleen tissue in the process of aging mice,and to analyze the biomarkers and their metabolic pathways closely related to 12-month-old male mice,thus to further explain the biological basis of the regulation effect of Jingui Shenqi pill. Methods: 10 male mice of 3-month-old were taken as the blank control,20 male mice of 12-month-old were randomly divided into the model group and the medication group; the medication group was treated with Jingui Shenqi decoction,whereas the blank control group and the model group were given the same volume of normal saline. Spleen tissues were collected for the study after 30 days. Results: There were 26 different endogenous metabolites related to growth and development,such as Xanthine,Pyruvic acid,Threonic acid and Glycocholic acid and so on,among which 11 were up-regulated,and 15 were down-regulated. Three metabolic pathways were involved. It found that Jingui Shenqi decoction had regulating effect on 9 biomarkers. Conclusion: Jingui Shenqi has intervention effect on aging changes in mice by regulating Chenodeoxycholic acid,Glycocholic acid and Xanthine.
