摘要
观察长期摄入二甲基亚硝胺(N-nitrosodimethylamine,NDMA)超标腊肠对大鼠肝脏的病理损害。通过饮用含30μg/kg NDMA饮用水建立大鼠肝脏损害模型,在NDMA造模组出现肝炎-肝纤维化-肝硬化-肝癌病理损害时,立即处理同期喂养的普通饲料对照组和腊肠喂养组大鼠,检测血清总蛋白(Total protein,TP)、白蛋白(Albumin,ALB)、天门冬氨基转移酶(Aspartate aminotransferase,AST)、丙氨酶氨基转移酶(Alanine aminotransferase,ALT)、碱性磷酸酶(Alkaline phosphatase,ALP),计算肝脏指数以及观察肝脏组织进行病理检查。结果显示,第7、17、25、33周时NDMA造模组大鼠肝脏出现典型肝炎、肝纤维化、肝硬化、肝癌病理改变,造模成功。长期腊肠摄入对大鼠一般状况以及肝脏指数影响不大,但对血清TP、ALB,AST、ALT、ALP水平有影响。整个实验期间(33周)肝脏病理改变从18%普通腊肠对照组(S1组)、18%腊肠NDMA超标组(S2组)至36%腊肠NDMA超标组(S3组)逐渐明显。各组肝炎评分从S1组→S2组→S3组→NDMA组逐渐增高,并随实验时间延长而增高。长期摄入NDMA污染腊肠导致大鼠肝脏损害,损害程度与腊肠中NDMA含量及喂养时间有关。
To observe the pathological changes of liver from rat following long -term intake of sausage withdifferent content of N-nitrosodimethylamine (NDMA) . Rat liver damage model was established by drinkingwater containing 30 μg/kg NDMA water. When the serial progression of this model was divided into theinflammation stage,the fibrosis stage,the cirrhosis stage,and hepatocellular carcinoma stage,the rats fromfeeding control diet group and sausage diet groups were executed. Serum total protein (TP),albumin(ALB),aspartate aminotransferase (AST),alanine aminotransferase (ALT)and alkaline phosphatase (ALP)weredetected. The liver index was calculated and the liver tissues were observed for pathological examination.Results showed that the pathological changes of typical hepatitis,liver fibrosis,cirrhosis and liver canceroccurred in the liver of NDMA model rats at the 7th,17th,25th and the 33th week,respectively. The generalcondition of rats in three sausage diet groups at different time points did not show any significant changescompared with control group,as did the change of liver index. But there are some changes in the level of serumTP,ALB,AST,ALT and ALP. The pathological changes of liver were gradually aggravated from 18 % controlsausage diet group (S1 group),containing 18 % NDMA exceeded the limit sausage diet group (S2 group)tocontaining 36 % NDMA exceeded the limit sausage diet group S3 group during the whole experimental period(33w) . The hepatitis score in each group continuously increased from S1 group,S2 group,S3 group to NDMAgroup,and increased with the extension of experimental time. The long-term intake of sausage with NDMA ex原ceeded limit led to rat liver damage,and the extent of damage was related to the content of NDMA of sausageand the feeding time in sausage diet.
