摘要
为建立花生根中白藜芦醇的高效液相色谱含量测定方法,采用HPLC法进行了试验,色谱条件为Shimpack C18色谱柱(150.0 mm×4.6 mm,5μm);流动相为甲醇-水(52∶48);检测波长306 nm;流速1.0 mL/min;进样量10μL;柱温30℃。结果表明,白藜芦醇进样量在0.2~1.0μg范围内与峰面积呈良好的线性关系,回归方程为Y=6 215.3X-65.647,r=0.999 8;平均回收率为99.21%;RSD为0.97%。该方法灵敏、简便、准确,可用于花生根的质量控制。
A resverse-phase high performance liquid chromatography (HPLC)method was developed to determine resveratrol content in peanut root.The separation was performed on the following condition :a Shimpack C18 column (150.0 mm ×4.6 mm, 5 μm), the mobile phase of methanol-water(52:48) ,flow rate was 1.0 mL/min and the detecfion wavelength was 306 nm,the injection volume was 10μL and column temperature was 30 ℃.The results showed that there was a good linear relationship between the area and the injection volume of resveratrol from 0.2 μg to 1.0 μg.The corresponding regression equation was Y=6 215.3X-65.647, r=0.999 8.The average recovery was 99.21% and RSD was 0.97%.The method is sensitive, accurate and speedy, therefore it is suitable for quality control of peanut root.
