烟草黑胫病拮抗菌的筛选鉴定及其发酵工艺优化 被引量:6

摘要 【目的】为筛选出对烟草黑胫病具有拮抗作用的细菌菌株,为烟草黑胫病的生物防治及生物农药的开发提供理论依据。【方法】采用平板对峙法筛选对烟草黑胫病有明显抑制作用的细菌菌株,结合形态观察、生理生化特征及分子生物学特征对该菌株进行鉴定,并利用菌饼法对该菌株的培养工艺进行优化。【结果】筛选出的菌株D1为革兰氏阳性菌、短杆状、产孢,且生理生化特征及16S rRNA基因序列与解淀粉芽孢杆菌具有极高的相似性(97%),因此确认菌株D1为解淀粉芽孢杆菌。筛选的最优培养基为PD液体培养基,最优碳源为蔗糖、氮源为蛋白胨、无机盐为氯化钙,最优配比为蔗糖20%、蛋白胨10%和CaCl_2 10%;最佳培养条件为pH 7~8、培养时间42 h和装瓶量30%。【结论】筛选得到的目的菌株通过发酵培养对烟草黑胫病有明显的抑制作用,可作为生防菌开发利用。 【Objective】The study aimed to screen bacterial strains with an antagonism against Phytophthora nicotianae and to provide the theoretical basis for biological control and bio-pesticide development of Phytophthora nicotianae.【Method】The bacterial strains with an obvious inhibition effect against Phytophthora nicotianae were screened by the plate confrontation culture method. The screened strain was identified by morphological observation,physiological and biochemical property,and biological characteristics. The culture process of the screened strain was optimized.【Result】The screened D1 strain was a Gram-positive bacterium with a short bacilliform and spore-production. The similarity in physiological and biochemical property and 16 S rRNA gene sequence between D1 stain and Bacillus amyloliquefaciens was up to 97%,which indicated that D1 strain was Bacillus amyloliquefaciens. The optimum medium for strain screening was PD + 20 g/L sucrose +10 g/L peptone + 10 g/L CaCl_2. The optimum culture conditions include p H 7-8,42 hours and 30 % medium volume( 100 m L/300 m L).【Conclusion】D1 strain with an obvious inhibition effect against Phytophthora nicotianae can be used for development and utilization of biological control bacteria.
