摘要
目的建立测定瑶药小钻中戈米辛R含量的反相高效液相色谱法。方法采用甲醇提取,应用高效液相色谱法(HPLC)对小钻中戈米辛R进行含量测定,色谱柱为Diamonsil C_18(4.6 mm×250.0 mm,5.0μm),流动相为甲醇-0.4%磷酸(含0.2%三乙胺)水梯度洗脱,检测波长254 nm,柱温30℃,流速0.8 m L/min。结果戈米辛R在0.380 8~4.188 8μg内呈良好线性关系,r=0.9999,戈米辛R平均回收率为97.19%,RSD=1.66%(n=9)。结论该方法简便,测定瑶药小钻中戈米辛R含量结果准确、可信,可有效控制小钻药材质量。
Objective To establish RP-HPLC method for content determination of gomisin R in Kadsura longipedunculata. Methods The methanol extraction was adopted and HPLC method was used detect the content of gomisin R in Kadsura longipedunculata. The separation was performed on a elite Diamonsil C_18 Column(250.0 mm × 4.6 mm,5.0 μm),the acetonitril-0.4% phosphoric acid and 0.2% triethylamine was used as the mobile phase,the detection wavelength was at 254 nm,the temperature was maintained at 30 ℃,the flow rate was 0.8 m L/min. Results The concent of gomisin R was linear within the range of 0.380 8-4.188 8 μg(r=0.999 9). The average recovery and RSD of the method were 97.19% and 1.66%(n=9). Conclusion The method is accurate,reproducible and can effectively be used to control the quality of gomisin R in Kadsura longipedunculata.
